摘  要:目的探讨秦皮乙素诱导人肝癌细胞SMMC-7721凋亡的分子机制。方法不同浓度秦皮乙素(1.12、2.24、4.48 mmol/L)处理人肝癌细胞SMMC-7721,流式细胞仪检测线粒体膜电位,分光光度法检测Caspase-3、Caspase-8、Caspase-9的活性,蛋白印迹法和免疫荧光法检测Bax、Bcl-2、Caspase-9蛋白的表达。结果秦皮乙素显著降低SMMC-7721细胞线粒体膜电位,提高Caspase-3、Caspase-9的活性,而对Caspase-8的活性无显著影响;升高Bax/Bcl-2比例,进而活化Caspase-9,并呈良好的剂量依赖关系。结论秦皮乙素可通过线粒体途径,启动下游的效应Caspase,诱导人肝癌细胞SMMC-7721凋亡。AIM To investigate molecular mechanism of human hepatoma cell SMMC-7721 apoptosis induced by aesculetin.METHODS Different concentrations of aesculetin(1.12 mmol/L,2.24 mmol/L,4.48 mmol/L) were used to treat human hepatoma cell SMMC-7721 and to monitor the changes of mitochondrial membrane potential with Flow Cytometry.The activities of Caspase-3,Caspase-8 and Caspase-9 were determined by spectrophotometric method.The protein expressions of Bax,Bcl-2 and Caspase-9 were studied by western blotting test and indirect immunofluorescence assay.RESULTS Aesculetin significantly reduced mitochondrial membrane potential of SMMC-7721,and increased activity of Caspase-3,Caspase-9.But there was no significant effect on the activity of Caspase-8.Aesculetin increased Bax/Bcl-2 ratio,activated Caspase-9,and showed good dose-dependent relationship.CONCLUSION Aesculetin starts the downstream effect of Caspase,and induces apoptosis of hepatoma cell SMMC-7721 through mitochondrial pathway.
