摘 要:目的建立高效液相色谱法测定大鼠血浆中绿原酸和木犀草苷。方法大鼠灌胃给予咽炎合剂(金银花、射干、山豆根、甘草和陈皮),血浆样品经叔丁基甲醚萃取,以槲皮素为内标,使用Phenomenex Luna C18色谱柱,流动相为0.4%醋酸溶液(A)-甲醇(B),B泵的比例0~40 min从10%上升到80%,检测波长350 nm;结果血浆中绿原酸在0.5~10 g/L、木犀草苷在0.1~2.5 g/L范围内与峰面积呈良好的线性关系。大鼠灌胃咽炎合剂后绿原酸和木犀草苷的呈药动学非房室模型。结论所建方法适合绿原酸和木犀草苷的药动学研究。AIM To establish an HPLC method for determination of chlorogenic acid and luteoloside contents in rat plasma.METHODS Yanyan Mixture(Lonicerae japonicae Flos,Belamcandae Rhizoma,Sophorae tonkinensis Radix et Rhizoma,Glycyrrhizae Radix et Rhizoma and Citri reticulatae Pericarpium) was administrated to rats by gastric lavage.After liquid-liquid extraction of the plasma by methyl t-butyl ether,the samples were analyzed on a Phenomenex Luna C18 column with mobile phase consisted of 0.4% acetic acid(A)-methanol(B),during the run time from 0 to 40 min,the phase B raised from 10% to 80%.The flow rate was 1.0 mL/min and the detection wavelength was set at 350 nm,using quercetin as internal standard.RESULTS For chlorogenic acid from 0.5-10 g/L,and for luteoloside from 0.1-2.5 g/L,the calibration curves showed good linear relationship with the peak areas.Chlorogenic acid and luteoloside showed non-compartment model after being administrated.CONCLUSION This method could be used for pharmacokinetics study of chlorogenic acid and luteoloside.
