摘  要:目的建立同时测定金匮肾气丸(山茱萸、牡丹皮、车前子、地黄等)中马钱苷、芍药苷、毛蕊花糖苷和丹皮酚的定量测定方法。方法采用HPLC法;Thermo Hypersil BDS C18色谱柱;乙腈-0.01%磷酸溶液为流动相;体积流量为1 mL/min;检测波长0~17 min,237 nm;18~25 min,233 nm;25~40 min,330 nm;40~55 min,275 nm。结果根据回归方程,4种成分线性范围分别为马钱苷0.075 6~0.945 5μg,r=0.999 7;芍药苷0.107 0~1.338 1μg,r=0.999 9;毛蕊花糖苷0.153 0~1.912 7μg,r=0.999 8;丹皮酚0.129 4~1.617 5μg,r=1.000 0;平均回收率分别为马钱苷99.2%,RSD为0.44%(n=6);芍药苷99.0%,RSD为0.84%(n=6);毛蕊花糖苷99.2%,RSD为0.37%(n=6);丹皮酚99.6%,RSD为0.28%(n=6)。结论该方法操作简便,准确、重复性好,可用于金匮肾气丸的质量控制。AIM To develop a RP-HPLC method for determining the contents of loganin,paeoniflorin,verbascoside and paeonol in Jinkui Shenqi Pills(Corni Fructus,Moutan Cortex,Plantaginis Semen,Rehamnniae Radix,etc.).METHODS The HPLC quantitative analysis was carried out on a column of Thermo Hypersil BDS C18 with a mobile phase of acetonitrile-0.01% phosphoric acid under a flow rate of 1 mL/min.The detection wavelengths were set at 237 nm(0-17 min),233 nm(18-25 min),330 nm(25-40 min),275 nm(40-55 min).RESULTS The linear ranges of loganin,paeoniflorin,verbascoside and paeonol fell within the ranges of 0.075 6-0.945 5 μg,0.107 0-1.338 1 μg,0.153 0-1.912 7 μg,0.129 4-1.617 5 μg,respectively.The four components showed good linear correlations(r=0.999 7,r=0.999 9,r=0.999 8,r=1.000 0).The recoveries were 99.2% for loganin,99.0% for paeoniflorin,99.2% for verbascoside and 99.6% for paeonol,respectively.The relative standard deviations were 0.44%,0.84%,0.37% and 0.28%,respectively(n=6).CONCLUSION This method is simple,accurate,reproducible and convenient for the quality control over Jinkui Shenqi Pills.
