摘 要:目的探讨重铬酸钾(K2Cr2O7)对雄性生殖系统的影响,为进一步研究K2Cr2O7的雄性生殖毒性机制提供理论依据。方法建立SD大鼠睾丸支持细胞(SC)的体外原代培养模型。利用噻唑蓝(MTT)比色试验测定K2Cr2O7对SC增殖活性的影响,确定染毒浓度。用逆转录聚合酶链式反应(RT-PCR)检测SC雄激素结合蛋白(ABP)、转铁蛋白(Tf)和抑制素(INH)mRNA的表达。结果ABP、Tf的mRNA相对表达量随K2Cr2O7浓度增加而升高,与对照组比较,ABP的mRNA相对表达量在K2Cr2O7浓度为10-7和10-6 mol/L时,Tf的mRNA相对表达量在K2Cr2O7浓度为10-6 mol/L时,差异有统计学意义(P<0.05)。INH的mRNA相对表达量随K2Cr2O7浓度升高而降低,在浓度为10-8 mol/L时,与对照组比较,差异有统计学意义(P<0.05)。结论K2Cr2O7能够引起大鼠SC分泌功能的改变,产生生殖毒性作用。Objective To explore the effects of potassium dichromate on male reproductive system and to provide theoretical criteria for further study of the mechanism of male reproductive toxicity induced by potassium dichromate. Methods Sertoli's cell (SC)of rat in primary culture models was established. The effect of potassium dichromate on activity of SC was determined by MTT colorimetric method. The concentration of potassium dichromate was confirmed by the activity measurement. The mRNA levels of ABP/Tf/INH were determined by real--time PCR. Results The relative expression of ABP/Tf's mRNA increased with the increase of the concentration of potassium dichromate. Compared with the control group, when the concentration of potassium dichromate was at 10-7 and 10-6 mol/L, the relative expression of ABP'S mRNA, and the relative expression of Tf's mRNA at the concentration of potassium dichromate 10-6 mol/L, the difference was significant(P〈0.05). The relative expression of INH's mRNA decreased with the decrease of the concentration of potassium dichromate. When the concentration of potassium dichromate was at 10 8 mol/L, the difference was significant in compare with the control group(P〈0.05). Conclusions Potassium dichromate could change the ability of secretory functions of rat Sertoli's cell, and produce the reproductive toxicity.
