摘 要:目的:探索羟丙基甲基纤维素(HPMC)作为水溶性药物的缓释骨架时,各处方因素对药物释放的影响。方法:以紫外分光光度法为检测方法、以累积释放度为检测指标,通过单因素实验,考察各处方因素对4种水溶性药物的HPMC骨架缓释片的药物释放的影响。结果:药物的溶解度、HPMC用量对药物释放的影响最为明显,其次是HPMC黏度;实验所选的填充剂种类和片剂硬度对药物释放的影响相对较小;阻滞剂用量、润滑剂(助流剂)种类、片剂表面积大小等因素的影响可以忽略。结论:研究结果有一定的规律,为水溶性药物HPMC骨架缓释片的研制提供一定的参考。Objective: To explore the influence of prescription factors on the release of water-soluble retard tablets with hydroxypropyl methylcellulose (HPMC) matrix. Method: In the single factor experiments, the cumulative release rate was detected by UV spectrophotometry to analyze the effects of various formulation factors to the release of four water-soluble retard tablets with HPMC matrix. Result: The amount of HPMC in the drug is the most important factor to influence the drug solubility ; the second important influencing factor is viscosity of HPMC, but the influence of bulking agent type and tablet hardness were weak. The influence of the block agent amount, the lubricant type or tablet surface area and size can be neglected. Conclusion: This article summarizes the law of retard tablets and provides a reference for the research of water-soluble retard tablets with HPMC matrix.
