摘  要:目的:探讨毛冬青总黄酮对大鼠血瘀合并脑缺血模型的影响。方法:采用大鼠连续注射地塞米松复制血瘀模型。毛冬青总黄酮混悬液(0.12,0.06,0.03 g·kg-1,20 mL·kg-1)连续灌服10 d,于第11天给药后1 h,夹闭大鼠双侧颈总动脉30 min造脑缺血模型。取血测全血黏度。断头取脑,一半脑置于10%福尔马林溶液中固定,用于HE染色;另一半脑用生理盐水制备脑匀浆,用于测定乳酸(LD)、乳酸脱氢酶(LDH)、三磷酸腺苷酶(ATP)酶水平以及丙二醛(MDA)含量。结果:血瘀合并脑缺血模型复制成功。与血瘀加脑缺血组相比,高、中剂量毛冬青总黄酮能显著降低模型大鼠全血黏度,高切为(7.25±1.78),(7.52±1.06)mPa.s,中切(8.75±2.02),(9.26±1.35)mPa.s,低切(17.06±3.77),(18.36±2.29)mPa.s,低剂量毛冬青总黄酮能显著降低全血黏度高切值(7.92±2.32)mPa.s,中剂量毛冬青总黄酮能显著降低模型大鼠脑匀浆中LD含量(3.51±0.69)mmol·g-1,显著提高LDH水平(1 822.2±128.0)U.mg-1,明显降低MDA含量(2.66±0.42)mmol·mg-1;高、中剂量毛冬青总黄酮能显著提高脑匀浆中Na+-K+-ATPase,Mg2+-ATPase,Ca2+-Mg2+-ATP活力(8.23±0.63),(8.53±0.52),(7.15±0.20),(7.98±0.24),(8.17±0.33),(8.54±0.29)μmol·mg-1·h-1,中剂量毛冬青总黄酮能显著提高Ca2+-ATPase活力(8.84±1.00)μmol·mg-1·h-1,小剂量毛冬青总黄酮能显著提高Na+-K+-ATPase活力(7.94±0.16)μmol·mg-1·h-1,各剂量组毛冬青总黄酮均可显著改善脑组织病理改变。结论:毛冬青总黄酮可显著改善大鼠血瘀合并脑缺血模型的脑缺血损伤。ObjeCtive: To explore the effect of the flavonoids of Ilex pubescens on blood stasis with / cerebral ischemia in rats. Method: Dexamethasone was used to induce blodd stasis model. Suspension of flavonoids of I. pubescens was given for 10 days, the carotid artery was blocked for 30 min 1 h after administration of determining on the day 11, Viscosity of whole blood was measured and brain tissue was collected for HE staining and determination of lactic acid ( LD ), lactate dehydrogenase ( LDH), adenosine triphosphate (ATP) enzyme levels and malondialdehyde (MDA) content. Result: The model of blood stasis with cerebral ischemia was successfully induced. Compared with model group the large and medium doses of the flavonoids of I. pubescens could reduce the viscosity of whole blood significantly, the value was (7.25 ± 1.78) , (7.52 ± 1.06), (8.75±2.02), (9.26 ±1.35), (17.06±3.77), (18.36±2.29) mPa.s. The small dose of the flavonoids of I. pubescens could reduce the whole blood viscosity, value being (7.92±2.32) mPa.s. The medium dose of the flavonoids of I. pubescens could reduce the LD content (3.51±0.69) mmol-g-1, increased LDH level (1 822.2 + 128.0) U.mg-1 significantly and lower MDA content (2.66± 0.42) nmol.mg-1, The large and medium dose of the flavonoids of L pubescens could significantly improve the Na + -K -ATPase, Mg2+ -ATPase, Ca2-Mg2-ATP activity, values were (8. 230 ±0. 633), (8.53±0.52), (7.15±0.20), (7.98±0.24), (8.17±0.33) , (8.54±0.29) p, mol .mg-1 .h-1. The medium dose of the flavonoids of I. pubescens could significantly improve the Ca2+-ATPase activity (8.84±1.00) μ molpi.mg-1- h-1. The small dose of the ilavonoids of I. pubescens could significantly improve the Na ~ -K + -ATPase activity (7.94± 0. 16) μmol-rag-1 h-1. Each group of the fiavonoids of L pubescens could significant improvement the pathological changes in brain tissue. Conclusion: The flavonoids of I. pubescens
