摘  要:目的:研究山茱萸提取物对黑素瘤细胞A375与角质形成细胞HaCaT共培养模型酪氨酸酶(TYR),酪氨酸酶相关蛋白-1(TRP-1)和酪氨酸酶相关蛋白-2(TRP-2)mRNA表达的影响。方法:以1∶2的比例构建的A375细胞与HaCat细胞共培养模型,将质量浓度为1.0,0.5 g·L-1的山茱萸提取物作用48 h,RT-PCR法检测药物对共培养模型中A375细胞TYR,TRP-1和TRP-2 mRNA表达。结果:与阴性对照组相比,山茱萸提取物1.0,0.5 g·L-1组TYR,TRP-1及TRP-2 mRNA表达量显著下调(P<0.01),mRNA下调率分别为36.07%,62.34%,44.69%和24.10%,53.00%,16.11%。结论:山茱萸提取物可能通过下调TYR,TRP-1和TRP-2 mRNA的表达而降低人黑素瘤细胞的黑素合成和酪氨酸酶活性。Objective: To observe the effects of the extract of Corni Fructus on the mRNA expression of tyrosinase (TYR) , tyrosinase related protein-1 ( TRP-1 ) and TRP-2 in co-culture model of melanoma cells and keratinocytes. Method : Differences concentration of the extract of Corni Fructus ( 1. 0, 0. 5 g . L- 1 ) were added in co-culture model of A375 melanoma cells and HaCat keratinocytes. RT-PCR assay was used to detect the effects of the extract of Corni Fructus on the TYR, TRP-1 and TRP-2 mRNA expression in A375 melanoma ceils. Result: Compared with control group, the extract of Corni Fructus ( 1.0, 0. 5 g.L-1 ) down-regulated mRNA expression of TYR, TRP-1 and TRP-2 in A375 melanoma cells by 36.07% , 62. 34% , 44.69% and 24. 10% , 53.00%, 16. 11% respectively (P 〈0,01 ). Conclusion: The extract of Corni Fructus can decrease the melanogenesis by down-regulating the mRNA expression of tyrosinase and its related protein.
