半枝莲多糖对肝癌细胞SMMC7721生长抑制及机制的研究

摘　　要：【目的】研究半枝莲多糖(Scutellaria barbata polysaccharides,SBP)对肝癌细胞SMMC7721生长的抑制及相关机制。【方法】将不同浓度SBP作用于SMMC7721肝癌细胞,采用四甲基偶氮唑盐(MTT)法检测半枝莲多糖对肝癌细胞SMMC7721生长增殖的抑制作用;实时荧光定量聚合酶链反应(Real-time PCR)检测细胞周期蛋白依赖性激酶(CDK2)与转录因子E2F1 mRNA表达;蛋白印迹法(Western blot)检测CDK2与E2F1蛋白表达。【结果】SBP对肝癌细胞SMMC7721具有明显的生长抑制作用,其中10、2.5μg/mL SBP组作用显著,均可使CDK2与E2F1 mRNA和蛋白表达下调。【结论】SBP对肝癌细胞SMMC7721具有增殖抑制作用,其机制可能与CDK2和E2F1表达下降有关。Objective To observe the effect of Scutellaria barbata polysaccharides （SBP） proliferation of hepatoma cell line SMMC7721 and to explore its relevant mechanism. Methods on inhibiting the SMMC7721 cells were cultured with different concentrations of SBP. The proliferation of SMMC7721 cells was detected by methyl thiazolyl tetrazolium （MTT） assay. The expression of CDK2 and E2F1 mRNA was detected by using real-time polymerase chain reaction （PCR）, and the expression of CDK2 and E2F1 protein was detected by using Western blot method. Results SBP could effectively inhibit the proliferation of SMMC7721 cells, and SBP at concentrations of 2.5 and 10μg · mL1 had obvious effect on decreasing the mRNA and protein levels of CDK2 and E2F1. Conclusion SBP can inhibit the proliferation of SMMC7721 cells, and its mechanism is possibly related with the decrease of CDK2 and E2F1 expression.
