仁术健胃颗粒对脾气虚证大鼠表达基质金属蛋白酶2的影响

摘　　要：目的探讨仁术健胃颗粒对慢性萎缩性胃炎(CAG)脾气虚证大鼠表达基质金属蛋白酶2(MMP-2)的影响。方法除正常组外,其他大鼠制备CAG脾气虚证模型。将模型大鼠随机分为5组:模型组(0.9%氯化钠溶液),胃复春组(胃复春0.86 g.kg-1),仁术健胃颗粒高、中、低剂量组(仁术健胃颗粒12,6,3 g.kg-1),均连续灌胃5周,正常组给予等量0.9%氯化钠溶液。实验第10周末处死大鼠,称脾脏质量,肉眼观察胃黏膜组织,快速免疫组化检测MMP-2表达。结果模型组胃黏膜呈深棕黄色,MMP-2表达高于正常组(P<0.01);与模型组(6.83±2.48)比较,仁术健胃颗粒高、中、低剂量组MMP-2[分别为(2.20±1.30),(1.20±0.45),(2.60±1.95)]均明显减弱(P<0.01或P<0.05),胃黏膜分别呈少量浅黄色、少量浅黄色和大量浅黄色。结论仁术健胃颗粒治疗慢性萎缩性胃炎与降低胃黏膜中MMP-2表达相关。Objective To investigate the effects of Renshujianwei granula on matrix metalloproteinase 2（MMP-2） in rats with spleen-qi deficiency syndrome chronic atrophy gastritis（CAG）.Methods Spleen-qi deficiency syndrome of the CAG model was established,except for the normal control.The model rats were randomly divided into the model control group（0.9% sodium chloride solution）,the weifuchun group（0.86 g·kg-1）,the high-dose（12 g·kg-1）,middle-dose（6 g·kg-1） and low-dose（3 g·kg-1） of Renshujianwei granula groups.Rats were i.g.treated for consecutive five weeks.All of the rats were sacrificed by the tenth weekend.The quality of spleen weighed,the gastric mucosa tissue was macroscopically observed and the expression of MMP-2 was detected by rapid immunohistochemistry.Results Gastric mucosa was deep claybank in the model group,in which the expression of MMP-2 was higher than that of the normal（P0.01）.In comparison to the model group,MMP-2 were significantly decreased in the high-dose,middle-dose and low-dose of Renshujianwei granula groups as（2.20±1.30）,（1.20±0.45）,（2.60±1.95）（P0.01 or P0.05）,respectively,and the gastric mucosa of which presented as a small amount light yellow,some yellowish,and large number yellow,respectively.Conclusion The therapeutic effect of Renshujianwei granula for CAG is associated with a reduction of MMP-2 expression in gastric mucosa.
