摘  要:目的研究TGF-β1沉默人大肠癌LoVo细胞对人巨噬细胞系28SC免疫功能的影响。方法将构建的TGF-β1siRNA稳定表达LoVo细胞与人巨噬细胞系28SC共培养,酶联免疫吸附实验检测培养液中IL-12和TNF-α水平,同时设立对照组空载体pSUPER-neo-gfp稳定转染LoVo细胞+28SC。结果 TGF-β1siRNALoVo+28SC组IL-12和TNF-α分泌量高于对照组(均P<0.05);TGF-β1+28SC组IL-12和TNF-α分泌量与LoVo+28SC组之间无明显差异(P>0.05)。结论大肠癌LoVo细胞分泌的TGF-β1能通过影响IL-12和TNF-α的分泌抑制巨噬细胞的免疫功能,有望通过阻断TGF-β1信号转导改善肿瘤免疫微环境抑制大肠癌的发生发展。Objective To study the effects of TGF-β1 silencing of human colon cancer LoVo cell on immune function of human macrophages cell line 28SC.Methods LoVo cells stably expressing TGF-β1 siRNA were cultivated together with human macrophages cell line 28SC.The levels of IL-12 and TNF-α in culture medium were detected by enzyme linked immunosorbent assay(ELISA).While a control group was established.Results The concentrations of IL-12 and TNF-α in TGF-β1siRNA LoVo+28SC group were higher than those in control group(P0.05).The concentrations of IL-12 and TNF-α in TGF-β1+28SC group and control group had no significant difference(P0.05).Conclusion TGF-β1 secreted by colon cancer LoVo cells can inhibit the immune function of macrophages through affecting the secretion of IL-12 and TNF-α.And it is hopeful to inhibit the development of colon cancer through blocking TGF-β1 signal transduction.
