摘 要:目的:探讨环孢素A(CsA)联合牙龈卟啉单胞菌脂多糖(Pg-LPS)局部用药对大鼠下颌第一磨牙颊侧牙龈面积及远中釉牙骨质界到牙槽脊顶距离(CEJ-ABC)的影响。方法:将40只大鼠分成8组,每组5只,各组大鼠下颌骨第一磨牙颊侧分别注射生理盐水,CsA(2.0mg.kg-1),高、中、低剂量的LPS(10.0,1.0,0.1μg)及CsA(2.0mg.kg-1)与不同剂量LPS的混合液。30d后处死大鼠,苏木精-伊红(HE)染色法处理大鼠下颌骨标本,观察大鼠下颌骨第一磨牙处牙龈组织、牙槽骨的变化。结果:CsA(2.0mg.kg-1)导致第一磨牙颊侧牙龈面积增加,远中CEJ-ABC距离变大,但与生理盐水组比较差异无统计学意义(P>0.05);高剂量的LPS(10.0μg)可以导致牙槽骨吸收,CEJ-ABC距离增加,高于生理盐水组(P<0.05);LPS(10.0μg)与CsA联合作用时对牙槽骨同样具有破坏作用,CEJ-ABC距离高于生理盐水组(P<0.05);中、低剂量LPS单独及与CsA联合应用时对牙槽骨均无明显影响,CEJ-ABC距离与生理盐水组比较均无明显差别(P>0.05);所有实验组的大鼠均未出现颊侧牙龈面积增大,与生理盐水组比较差异无统计学意义(P>0.05)。结论:低剂量CsA(2.0mg.kg-1)局部应用于大鼠牙周组织,既不引起牙龈增生,也不引起牙槽骨的吸收;低剂量CsA对高剂量LPS(10.0μg)引起的牙槽骨吸收无明显的逆转作用。Objective To explore the effects of cyclosporine A(CsA) and porphyromonas gingivalis-lipopolysaccharide(Pg-LPS) on the area of buccal gingiva of the first molar of mandibular and CEJ-ABC distance of rats.Methods Forty rats were equally divided into eight groups.There were 5 rats in each group.The first molar of mandibular of rats in various groups were injected,from the buccal side,with saline,CsA(2.0 mg·kg-1),LPS(10.0,1.0,0.1 μg),CsA+ LPS(10.0 μg),CsA+ LPS(1.0 μg),CsA+ LPS(0.1 μg),respectively.After 30 d,the mandibles were then dissected and sectioned for histological and immunocytochemical analysis by haematoxylin and eosin staining,the changes of the distance of CEJ-ABC and the area of buccal gingiva in various groups were observed.Results CsA(2.0 mg·kg-1) increased the area of the buccal gum of first molar and the distance of CEJ-ABC,but there was no statistically significant difference compared with control group(P0.05).LPS(10.0 μg) alone or combined with CsA could damage the alveolar bone and increase the distance of CEJ-ABC,and there were statistically significant differences compared with control group(P0.05);but there was no significant difference when treated with LPS(1.0,0.1 μg) alone or combined with CsA;and no obvious change in gingival was found following all of the treatments(P0.05).Conclusion Low dose of CsA(2.0 mg·kg-1) has no damage on the periodontal tissues of rats,and such a level of CsA may not decrease the periodontal breakdown induced by high dose of LPS(10.0 μg).
