卵泡中肝素结合表皮生长因子样生长因子的表达及其与卵泡发育的关系

摘　　要：目的探讨超促排卵过程中产生的卵泡中是否存在肝素结合表皮生长因子样生长因子(HB-EGF)的表达及其与卵泡发育的关系。方法选择42例接受体外受精—胚胎移植治疗不孕症的患者,超促排卵结束穿刺取卵时收集废弃的卵泡液及黄素化颗粒细胞,采用Western blot分别检测卵泡液中溶解型HB-EGF(sHB-EGF)和颗粒细胞中的膜结合型HB-EGF(proHB-EGF)。结果所有卵泡中均检测到sHB-EGF和proHB-EGF蛋白;同一卵泡的卵泡液中sHB-EGF含量明显大于颗粒细胞中proHB-EGF的表达量(P<0.05);大卵泡卵泡液中sHB-EGF和颗粒细胞中proHB-EG含量明显大于小卵泡(P<0.05);但小卵泡中sHB-EGF与proHB-EGF比值明显高于大卵泡。结论超促排卵过程中产生的卵泡中存在HB-EGF的表达,其表达量与卵泡的发育成熟有关。Objective To investigate whether heparin-binding epidermal growth factor-like growth factor （HB-EGF） is expressed in ovarian follicles formed by controlled ovarian stimulation （COS） with gonadotropins in the in vitro fertilization-embryo transfer （IVF-ET） program, and explore relationship between the HB-EGF content and follicular development. Methods Follicular fluid and the trans-membrane pre- cursor form of HB-EGF （proHB-EGF） in the luteinized granulosa cells of the follicles were collected at the time of oocyte retrieval in file IVF-ET program. Western blot was carried out to measure the protein content of soluble HB-EGF （sHB-EGF）. Results sHB-EGF and proHB-EGF were observed in both follicular fluid and lunteinized granulosa cells of all the follicles. In the same follicle, the amount of sHB- EGF was significantly higher than proHB-EGF （P 〈 0.05）. The amount of sHB-EGF and proHB-EGF in the large follicles （〉16 mm） was significantly higher than that in the smalle follicles （〈14 ram）. However,the ratios of sHB-EGF to pmHB-EGF of the small follicles were markedly higher than that of the large folhcles. Conclusion Both sHB-EGF and proHB-EGF are present in the ovarian follicles formed af- ter COS. The amount of their expression may be associated with the follicular growth and maturation.
