摘 要:目的:研究苦参碱对儿茶酚胺(CA)诱发豚鼠左心室流出道自律细胞电生理异常及苦参碱抗心律失常的干预作用。方法:应用常规细胞内玻璃微电极记录手段,观察正常灌流液组;肾上腺素(E,80μmol/L)、去甲肾上腺素(NE,80μmol/L)、异丙肾上腺素(Iso,80μmol/L)灌流液组;和肾上腺素(E)、去甲肾上腺素(NE)、异丙肾上腺素(Iso)灌流液分别+苦参碱(100μmol/L)组灌流液对豚鼠左心室流出道慢反应自律细胞,动作电位0相幅值(APA),细胞最大舒张电位(MDP),舒张期除极速率(VDD),4相自动去极速度(Vmax),50%复极化时间(APD50),80%复极化时间(APD80),自发放电频率(RPF)的影响。结果:与正常对照组对照,儿茶酚胺灌流液组左心室流出道慢反应自律细胞20min后APD50均明显缩短(P<0.01),RPF、VDD显著变快(E、Iso、P<0.01;NE、P<0.05);APA增大,Vmax明显加快。在不同儿茶酚胺灌流液组中加入苦参碱可明显延缓APD50、APD80;并使APA减小、Vmax减慢、RPF逐渐变慢,稳定20min后,自发放电频率(RPF)基本恢复正常节律。结论:儿茶酚胺类物质可明显影响豚鼠左心室流出道慢反应自律细胞电活动,使其自律性增加,而苦参碱能减弱儿茶酚胺的影响作用,降低诱发所导致的自发放电频率的升高,提示苦参碱可阻断儿茶酚胺诱发的左心室流出道慢反应自律细胞异常电生理所致的心律失常效应。Objective: To study the matrine on catecholamine(CA) induced guinea pig left ventricular outflow tract pacemaker cells electrophysiology anomaly and matrine antiarrhythmic intervention effect.Methods: by using conventional cells glass microelectrode recording means,observe normally perfusate group;Epinephrine(E,80 μ mol/L),Norepinephrine(NE,80 μ mol/L),Isoproterenol(Iso,80 μ mol/L) perfusate group;And epinephrine(E),norepinephrine(NE),isoproterenol(Iso) perfusion fluid respectively + matrine(100 μ mol/L) group perfusate on guinea pig left ventricular outflow tract slow reaction pacemaker cells,amplitude of action potential(APA),cell maximum diastolic potential(MDP),velocity of diastolic depolarization(VDD),maximal rate of depolarization(Vmax),50% and 80% of action potential duration(APD50 and APD80),rate of pacemaker firing(RPF).Results: compared with normal control group comparison,catecholamine perfusate group of left ventricular outflow tract slow reaction pacemaker cells after 20 min APD50were significantly shortened(P0.01),RPF,VDD significant increased(E,Iso,P0.01;NE,P0.05);APA increased,Vmax significantly increased.In different catecholamine perfusate group to join matrine can obviously shortened the APD50,APD80;And make APA decrease,Vmax slow,RPF gradually slow,steady 20 min,RPF recover normal rhythm.Conclusion: catecholamine can significantly affect the guinea pig left ventricular outflow tract slow reaction pacemaker cell electrical activity,make its autorhythmicity increases,and matrine can weaken the effect of catecholamine,reduce induced caused by the spontaneous discharge frequency increases,Matrine can block the effects of CA.
