摘 要:目的探讨高速逆流色谱法(HSCCC)分离丁公藤中东莨菪内酯的可行性及工艺。方法用纯甲醇超声萃取三次,得到丁公藤粗提物,再用HSCCC技术分离得到高纯度的东莨菪内酯。结果正己烷-乙酸乙酯-甲醇-水溶剂系统(体积比为3.0∶5.0∶3.5∶5.0)具有较好的分离效果;较佳工艺条件为:转速850 r/min、流动相流速2 mL/min和水浴温度25℃。结论采用HSCCC技术分离得到的东莨菪内酯样品经高效液相色谱法检测,纯度可达98.04%。Objective To study the feasibility and techniques for isolating scopoletin from Erycibe obtusifolia by high-speed counter-current chromatography(HSCCC).Methods Erycibe obtusifolia was ultrasonically extracted in 100% methanol three times,and then a HSCCC unit was used to get high purity of scopoletin.Results The preferable solvent system was composed of hexane-ethyl acetate-methanol-water(3.0∶50∶3.5∶5.0,V/V),and the preferable technical conditions were as followed: the rotate speed was 850 r/min,the mobile phase flow rate was 2 mL/min,and the bath temperature was 25℃.Conclusion The purity of scopoletin sample isolated by HSCCC is 98.04% through the technique of high performance liquid chromatography(HPLC).
