摘  要:为了解河南省猪群中猪流感流行状况,对河南多个地区的63头病死猪进行SIV分离鉴定,并对获得的分离株进行生物学特性研究.从病死猪的肺脏中分离到3株H1N2亚型SIV,分别命名为:A/Swine/HeNan/01/2010(H1N2)、A/Swine/HeNan/02/2010(H1N2)和A/Swine/HeNan/03/2010(H1N2).其中,A/Swine/HeNan/01/2010(H1N2)和A/Swine/HeNan/02/2010(H1N2)均能凝集鸡、小鼠、兔、猪、豚鼠、绵羊和牛的红细胞,均为热稳定型,其中前者EID50和ELD50较高,分别为10-7.16和10-6.54;后者较低,分别为10-5.86和10-3.83.A/Swine/HeNan/03/2010(H1N2)对猪和牛的红细胞无凝集活性,为中等热稳定型,且EID50和ELD50较低,分别为10-2.43和10-2.67.同时3株SIV均对氯仿、乙醚和酸敏感,均可引起MDCK细胞病变,但不能感染BALB/c小鼠.In order to understand the prevalence of swine influenza in Henan Province, 63 samples of lungs from dead pigs were collected from 11 cities of Henan and the biological characteristics of swine influenza virus were analysed. The results showed that the 3 swine influenza viruses isolated were all H1 N2 subtypes, named A/Swine/HeNan/ 01/2010 ( HI N2 ) , A/ Swine/HeNan/02/2010 (H1N2) and A/Swine/HeNan/03/2010(H1N2), respectively. On this basis, these isolation strains were pu-rifieated by limited dilution cloning. Biological characteristics research showed: Except that A/Swine/ HeNan/03/2010(H1N2) could not agglutinate the pig and cattle erythrocytes, the remained two strains could agglutinate erythrocytes of chicken, sheep, Guinea pig, rabbit, mouse, pig and cattle; Hemagglutinin thermostability test results displayed A/Swine/He Nan/01/2010 (H1N2) and A/ Swine/HeNan/02/ 2010 (H1N2) belonged to thermstabile type, and A/ Swine/HeNan/03/2010 ( H 1 N2) belonged to mid-thermstabile type. A/Swine/HeNan/01/2010 ( H 1 N2 ) showed higher path-ogenicity to chicken embryo, of which EID50 and ELD50 were 10^-7.16 and 10^-6.54, respectively, and the others were lower. All the 3 strains were sensitive to ether, chloroform and acid and showed infectivity cytopathic effect in the MDCK cell, but they could not infect Balb/c mice in this study.
