摘 要:对我国口岸截获的5种乳白蚁及黄胸散白蚁的ITS区基因序列进行克隆并测序,进行序列分析和构建系统发育树,探讨其系统发育关系。研究结果表明:乳白蚁rDNA-ITS基因序列长度约900 bp,CG碱基含量为62.3%,明显高于AT含量(37.7%)。同巢群3种不同品级的乳白蚁(工蚁、兵蚁、有翅成虫)ITS序列没有遗传分歧。乳白蚁属内不同种间的同源性为94.3%~98.7%,同种不同地理种群之间的同源性均大于99%。大家白蚁C.curvignathus和塞庞乳白蚁C.sepangensis关系最近,婆罗乳白蚁C.travians与其他种进化距离最远。Phylogenetic and sequence analyses of ITS gene were conducted to elucidate the relationships among 5 species of Coptotermes, which intercepted by CIQ or collected from several geographical of China. The length of rDNA-ITS gene of Coptotermes was about 900 bp, and the content proportion of ITS sequence of CG reached to 62.3%, higher than AT content proportion(37.7%).Three castes from one clone of Coptotermes shared identical ITS sequence. Intraspecifical and interspecifical identity percent of Coptotermes ranged from 94.3%-98.7% and 99%, respectively. Phylogenetic analysis showed a close relation between C. curvignathus and C. sepangensis while distant relation between C. travians and other species.
