摘 要:在对 10 5份湖北栽培大麦酯酶同工酶研究的基础上 ,采用 RAPD技术对分属于 8种酶谱类型的 10份湖北栽培大麦品种进行了研究 .6 3个随机引物中有 12个引物产生稳定多态性的带 ,共扩增出 73条带 ,其中 46条表现出多态性 .将每一条带视为一个独立的性状 ,按其有无列出二元数据矩阵 ,计算 Nei氏相似性系数 (S)和遗传距离 (D) ,列出遗传距离矩阵 ,采用 PHYL IP3.5 c软件包进行聚类 .讨论了 RAPD的稳定性以及 RAPD标记与同工酶标记的异同 .Based on the previous study on the esterase isozyme zymogram of 105 Hubei barley, 10 materials of which belonging to the eight zymogram patterns respectively were investigated by using randomly amplified DNA (RAPD) method. 12 of 63 random primers produced 73 bands in total, and 46 bands of which showed polymorphisim. Each amplified band was looked as an independent character and scored for the presence or absence in a binary matrix. Nei's similarity coefficient (S) and genetic distance (D) were calculated. PHYLIP 3.5c was used to generate a dendrogram. The genetic relationships among barley tested were investigated and the difference between cluster analysis of isozyme zymogram and that of RAPD markers was compared. Factors affecting the stability of RAPD profiles were discussed also.
