摘 要:目的建立LC-MS/MS法测定人血浆中盐酸非那吡啶浓度。方法色谱柱为Capcell PAK C18,流动相为甲醇-10 mmol乙酸铵(85∶15,pH=4),流速为0.15 mL.min-1,电喷雾离子源(ESI),正离子方式检测,多反应监测(MRM)。结果盐酸非那吡啶的线性范围为0.2~50.0 ng.mL-1,日内、间差RSD均≤11.5%,提取回收率为≥60.2%。结论该方法灵敏、准确、快速,可用于人血浆盐酸非那吡啶浓度的测定。Objective To establish a new LC–MS/MS method for the determination of phenazopyridine hydrochloride in human plasma.Methods The analysis column was conducted with Capcell PAK C18.methanol-ammonium acetate(5∶15) was served as mobile phase.The flow rate was 0.15 mL·min-1.Detection was performed on a tandem mass spectrometry with electrospray ionization source.Multiple reactions monitoring mode was used to quantify phenazopyridine hydrochloride.Results The linear calibration curves were obtained in the range of 0.2-50.0 ng·mL-1for phenazopyridine hydrochloride.The extraction recovery for phenazopyridine hydrochloride were≥60.2%.The intra and inter day precision were≤11.5%.Conclusion The method is accurate,sensitive,and convenient,suitable for therapeutic drug monitoring and pharmacokinetic study of phenazopyridine hydrochloride.
