摘  要:目的用高效液相色谱法追踪氯氮平片的体内代谢过程。方法分别采集60例患者的胃液、血浆、粪便与尿液,滤膜过滤后,用乙醚提取、挥干,以流动相溶解残渣,测定其中的氯氮平浓度并观察其分解产物。结果液相方法回收率在94.0%以上,相对标准偏差<2%,氯氮平在0.05~100.00μg.mL-1呈良好线性关系,定量下限为7.54 ng.mL-1。60例患者中,44例患者显效,16例未见好转。结论该法便捷灵敏,可用于氯氮平片的体内代谢分析。Objective To track the catabolic route of clozapine tablets in vivo by HPLC.Methods The gastric juice,plasma,feces and urine of 60 patient exemplars was collected,filtrated by membrane and extracted with ether.Then the solvent was evaporated and the residue was dissolved in the mobile phase,in which the concentration of clozapine was assayed and its catabolites were analyzed.Results The methodical recovery was above 94.0%.The relevant standard deviation was within 2%.Clozapine was in good linear at the concentration of 0.05~100.00 μg·mL-1,and the minimum limit of quantification was 7.54 ng·mL-1.Clozapine was curative effective to 44 exemplars,while 16 exemplars did not respond to its clinical effect.Conclusion The method is proved to be prompt and sensitive,appropriate to the catabolic analysis in vivo and clinical evaluation to clozapine tablets.
