摘 要:根据油茶籽多糖Ⅰ所拥有的特异性磷酸基团,运用固化金属离子亲和层析法获得合适的金属螯合剂,使得具有一定降血脂作用的油茶籽多糖Ⅰ能特异性的被结合到琼脂糖凝胶上,从而获得纯度较高的油茶籽多糖Ⅰ。通过试验对亲和层析法在提取过程中的使用条件进行优化,得到最适pH以及离子类型。此外,试验将传统方法与亲和层析法进行比较,其对油茶籽多糖Ⅰ的纯化效果,尤其在去蛋白方面,后者能更有效地去除油茶籽多糖Ⅰ里残留的蛋白质。The experiment was based on specific phosphate groups of oil tea seed polysaccharide I , the use of solidifying metal ion affinity chromatography to obtain a suitable metal chelating agents, so that the oil a lipid - lowering effect of tea seed polysaccharide I can be integrated into specific agar glucose gel, to obtain high purity of the oil tea seed polysaccharide I . Through the experiments, the affinity chromatography method in the extraction process of the conditions of use was optimized, getting the optimal pH and ion type. In addition, the experiment compared the traditional method with the affinity chromatography method, the purification effect of oil tea seed polysaceharide I , especially in the protein removing, the latter can more effectively remove the residual protein in oil tea seed polysaccharide I.
