siRNA沉默PRL-3基因对前列腺癌PC-3细胞生物学行为的作用

摘　　要：目的探讨小干扰RNA沉默PRL-3基因对前列腺癌PC-3细胞侵袭及增殖的影响。方法化学合成PRL-3基因的特异性siRNA,脂质体转染法转染PC-3细胞;24及48小时行real-time PCR和Western blotting验证其沉默效果;Tran-swell侵袭实验检测其体外侵袭力的改变;CCK8检测其增殖能力。结果转染siRNA后,与对照组相比较,siRNA干扰组mRNA水平抑制率达60%以上,蛋白水平抑制率达90%以上,且在48h达到最佳抑制效果(P<0.05,P<0.05);PC-3细胞穿过基膜数明显减少(P<0.05),但是对细胞生长抑制作用不明显。结论应用RNA干扰技术沉默人前列腺癌PC-3细胞中的PRL-3基因,可以抑制PC-3细胞的侵袭能力,为进一步探讨前列腺癌的发病机制奠定一定的实验基础。Objective To investigate the effects of small interfering RNA （siRNA） on the invasion and proliferation of PRL - 3 in human prostate cancer cell line PC - 3 ceils. Methods The siRNA for PRL - 3 gene was transfected into PC - 3 cells by Lipo- fectamine2000. The 24 hours and 48 hours silencing effects of PRL - 3 gene was assessed by real - time PCR and Western - blotting. The invasion of PC - 3 cells was analyzed by Transwell assay. The proliferation of PC - 3 cells was analyzed by CCK8 assay. Results After the siRNA for PRL -3 gene was transfected into PC -3 ceils, the expression of mRNA and protein were both decreased （P 〈 0.05 ）and at 48 hours reached the best inhibition effect. The invasive abilities were reduced obviously（ P 〈 0.05 ） , but for cells growth the effect was small （P 〉 0.05）. Conclusion Applicating the siRNA to silence PRL - 3 gene can inhibit the invasive ability of PC - 3 cells, and laid the experimental basis to further explore the pathogenesis of prostate cancer.
