摘  要:目的通过检测新生鼠肠损伤时回肠组织二胺氧化酶(diamine oxidase,DAO)和E-钙黏蛋白(E-cadherin,E-cad)的动态变化,探讨其在新生儿坏死性小肠结肠炎(necrotizing enterocolitis,NEC)中的作用。方法 48只健康1日龄内新生鼠,随机分为对照组和实验组,实验组腹腔注射5mg/kgE coli O55:B5内毒素(LPS),于LPS注射后1、3、6、12和24h随机进行动物处死,分离胃肠道,部分回肠组织用于形态学观察,其余用于DAO活性、E-cad mRNA测定。结果实验组大鼠腹腔注射LPS后1、3、6、12和24h回肠组织损伤评分分别为1.28±0.62,1.75±0.74,1.98±0.75,2.85±0.41和2.35±0.63,均明显高于对照组0.12±0.17(P<0.05);1、3、6、12和24h DAO活性变化分别为19.02±4.25,14.09±2.3,10.57±2.16,9.55±2.49和11.86±5.54U/mg,3、6、12和24h显著低于对照组20.83±3.55U/mg(P<0.05),DAO活性与肠损伤程度呈明显负相关(γ=-0.943,P<0.05);LPS注射后E-cad mRNA表达下调,E-cad mRNA与肠损伤程度无明显相关性(γ=0.657,P>0.05)。结论肠黏膜DAO活性可间接反映肠黏膜的完整性及损伤程度;E-cad不但与肠黏膜损伤有关,可能还参与了肠黏膜屏障的修复机制。Objective To study the content changes of diamine oxidase(DAO) and E - cadherin(E - cad) in neonatal rat model of intestinal injury, and analyze the function of DAO and E - cad in necrotizing enterocolitis. Methods Totally 48 neonatal rats of one day old were randomly divided into test group and control group. Test group were induced by intraperitoneal injection of LPS(5 mg/kg, E coli O55 :B5 ) and were put to death at 1,3,6,12,24 hours after LPS administration, then the intestine around ileocecal junction were taken. Pathological damages of ileocecal tissues were checked with light microscope. DAO and E - cad levels were determined by reverse tran- scription polymerase chain reaction( RT- PCR). Results The scores of histological evaluation in test group which sacrificed at 1,3,6, 12,24 hours were ( 1.28±0.62 ) , ( 1.75±0.74 ) , ( 1.98 ±0.75 ) , ( 2.85±0.41 ), ( 2.35 ±0.63 ) respectively and higher than control group ( 0.12±0.17 ) ( P 〈 0.05 ). The activity of DAO in test group which sacrificed at 1,3,6,12,24 hours were 19.02 ± 4.25,14.09± 2.3,10.57±2.16,9.55±2.49 and 11.86±5.54U/mg, respectively. The activity of DAO in control group were 20.83±3.55U/mg and higher than test group which sacrificed at 3,6,12,24 hours(P 〈 0.05). The relation of the degree of intestinal injury and the activity of DAO in intestinal tissue was negative(γ = -0. 943, P 〈 0.05 ). The level of E -cad mRNA went down in test group which induced by intraperitoneal injection of LPS. The expression of E - cad mRNA does not correlate with degree of intestinal injury (γ = 0. 657, P 〉 0.05 ). Conclusion The activity of DAO can be considered as studying the degree of intestinal injury. The expression of E - cad mRNA not only correlates to the degree of intestinal injury, but also probably benefits on repairing of intestinal barrier.
