摘  要:目的研究不同浓度氟对大鼠丙二醛(MDA)和谷胱甘肽过氧化物酶(GSH-Px)的活性及其mRNA表达的影响。方法在饮水中加入不同剂量的氟化钠喂饲大鼠1月后,测定血清氟含量、尿氟含量、肝组织MDA含量、GSH-Px的活性及其mRNA的表达,并进行相关分析。结果随染氟剂量的增加,1月后各剂量组大鼠血氟浓度相应增加,存在剂量-效应关系(r=0.884,P<0.01),但各剂量组间血氟浓度差异无统计学意义;各剂量组大鼠尿氟浓度相应增加,存在剂量-效应关系(r=0.921,P<0.01);各剂量组大鼠MDA含量升高,与对照组比较有统计学意义(P<0.01),存在剂量-效应关系(r=0.797,P<0.01);各剂量组大鼠GSH-Px的活性下降,与对照组比较有统计学意义(P<0.01),存在剂量-效应关系(r=-0.912,P<0.01);各剂量组大鼠GSH-Px的mRNA表达下降,与对照组比较有统计学意义(P<0.05),存在剂量-效应关系(r=-0.909,P<0.01)。结论氟中毒诱导大鼠肝组织GSH-Px的mRNA表达下降,可能是氟致脂质过氧化反应的机制之一。Objective To study the effect of different dosages of fluoride on the content of malondialdehyde(MDA) and activity and mRNA expression of glutathion peroxidase(GSH-Px) in rat liver.Methods Rats were given drinking water with various dosages of sodium fluoride for 1 month.The levels of fluoride in serum and urine,the content of MDA and the activity and mRNA expression of GSH-Px liver in rat liver were measured.Results The serum and urine fluoride and the content of MDA were significantly increased in dose-effect manner with the increased fluoride exposure.However,no significant difference of serum fluoride among different dose groups was seen.In addition,the activity and mRNA expression of GSH-Px in rat liver were decreased in dose-dependent manner.Conclusion Fluoride exposure induce the decreased mRNA expression of GSH-Px,which might be one of mechanisms underlying fluoride-induced lipid peroxidation.
