摘  要:目的探讨DcR3在类风湿性关节炎(RA)发病过程中表达水平的变化及其与疾病活动性关系。方法选择39例RA患者为实验组,26例健康体检者为对照组,采用实时荧光定量PCR(FQ-PCR)检测RA患者外周血单个核细胞DcR3mRNA的基因表达水平,采用ELISA法检测患者血清中DcR3、IL-4和IFN-γ水平,并将检测结果与DAS28评分进行相关性分析。结果活动期RA患者PBMC中DcR3mRNA表达水平高于缓解期RA患者和健康对照组,差异有显著性(P<0.05);活动期RA患者血清DcR3和IFN-γ水平显著高于缓解期RA患者和健康对照组(P均<0.01);IFN-γ/IL-4比值显示,活动期RA患者显著高于健康对照组和缓解期RA患者(P均<0.01);RA患者外周血DcR3mRNA水平和血清IFN-γ/IL-4比值与DAS28评分呈显著正相关趋势。结论活动期RA患者外周血DcR3蛋白与PBMC中DcR3mRNA表达水平显著升高,可能通过促Th2等作用参与RA的发生发展,与疾病的活动性密切相关。Objective To investigate DcR3 Levels of peripheral blood in rheumatoid arthritis(RA),and analyzes the correlation between the DcR3 Levels of Peripheral Blood and the Disease Activity in Rheumatoid Arthritis Patient.Methods The expression levels of DcR3 mRNA in peripheral Blood monocytes of 39 RA patients and 26 healthy controls were detected by real-time polymerase chain reaction(FQ-PCR),the levels of DcR3、IL-4 and IFN-γ in serum were detected by ELISA(enzyme-linked immuno sorbent assay).Results The expression levels of DcR3 mRNA in active stage of RA patients were higher than that of remission stage of RA patients and healthy controls(P0.05);The levels of DcR3、IFN-γ and IFN-γ/IL-4 in active stage of RA patients were higher than that of remission stage of RA patients and healthy controls(P0.01);The correlationships between DcR3 mRNA levels、IFN-γ/IL-4 and active renal score(DAS28)show positive correlation.Conclusion The expression levels of peripheral blood DcR3 were significant increase in RA active patients and closely related with the activity of the disease which suggesting that DcR3 migh involved in the pathological process.
