摘  要:目的探讨同型半胱氨酸(Hcy)干扰细胞周期素A(cycfinA)表达进而扰乱细胞增殖周期之后对平滑肌细胞(VSMCs)增殖的影响。方法正常人脐动脉VSMCs体外培养,随机分为对照组、叶酸治疗组(Hcy100μ M+叶酸30μM)、Hcy干预组(50、100、200、500μM)总计6组,分别以MTT检测VSMCs的增殖率.流式细胞仪测定细胞周期,荧光定量PCR测定CyclinA基因mRNA水平。结果MTT结果显示,不同浓度Hcy干预VSMCs72h后,Hcv对VSMCs细胞增殖的影响与剂量相关,随Hcy浓度增加细胞增殖显著,与对照组比较差异有统计学意义(P〈0.01)。流式细胞仪检测VSMCs周期分布结果显示,对照组细胞大多数处于G0/G1期,随着Hcy浓度的增加,VSMCs处于S期的细胞呈剂量依赖性增加,与对照组比较,差异有统计学意K(P〈001)。荧光定量PCR结果显示,不同浓度的Hcy干预VSMCs72h后,干预组与对照组相比CyclinA基因mRNA随Hcv浓度的增加而增加,叶酸治疗组与100μM Hcy组比较cyclinA mRNA表达降低(P〈0.05)。结论Hcy可通过调节CyclinA mRNA水平.影响细胞周期各时相分布途径,使得VSMCs的细胞周期调控紊乱,促进VSMCs的增殖,且叶酸可以拮抗Hcy引起的平滑肌细胞增殖的作用。Objective To explore homocysteine (Hcy) interference cyclinA expression then disrupt the cell proliferation cycle of vascular smooth muscle cells (VSMCs) proliferation. Medthods Cultured VSMCs were divided into control group, treated group (Hcy 1001μM+folicacid 301μM) and different concentrations of Hcy group (50, 100, 200, 500lμM) at random. The proliferative rate of VSMCs was detected by MTT assay and cell cycle distribution was determined by flow cytometer. The expression of CyclinA mRNA was examined by qPCR. Results The results of MTT assay showed that different concentrations of Hcy on the intervention VSMCs after 72h, with Hcy concentrations increasing, the multiplication rate of the VSMCs increased to compare with the control group (P〈0.05). Flow cytometry detecting VSMCs cycle distribution that the results showed that most of the control group cells belonged to the G0/G1 phase with Hcy concentrations increasing, VSMCs in S phase in a dose-dependent increased, it was significant difference (P〈0.05) to compare with control group, qPCR results showed that different concentrations of Hcy on VSMCs for 72h, with Hcy concentrations increasing, the levels of Cyclin A mRNA were increased to compare with the control group (P〈0.05). Conclusion Hcy could promote the proliferation of VSMCs through affecting the expression of Cyclin A which made the disorder of the VSMCs in cell cycle regulation. Folic acid bad the antagonistic effects on proliferation of VSMCs which induced by Hcy.
