摘 要:目的:建立嗅球摘除抑郁大鼠模型,并观察逍遥散对模型动物行为学指标和脑内单胺类神经递质含量的影响。方法:手术摘除方法建立SD大鼠的嗅球摘除抑郁模型。逍遥散高、中、低剂量(30g/kg、15g/kg、7.5g/kg)和阿米替林(0.01g/kg)连续灌胃给药4周,测定模型大鼠体重、旷场活动量及糖水消耗量,Morris法观察大鼠学习记忆能力,ELISA法测定大鼠脑内海马和皮质部位5-羟色胺(5-HT)、多巴胺(DA)水平。结果:与正常组比较,嗅球摘除抑郁模型大鼠体重下降、敞箱活动中水平与垂直活动均增加、糖水消耗量降低(P<0.05或P<0.01);水迷宫实验中定位航行潜伏期明显延长(P<0.01),空间探索实验中穿越平台的次数显著降低(P<0.05);海马部位5-HT含量明显下降(P<0.05)。与模型组比较,逍遥散高、中、低剂量(30g/kg、15g/kg、7.5g/kg)和阿米替林(0.01g/kg)组对模型动物体重的降低具有一定对抗作用,连续给药4周能减少模型动物站立次数或穿越格子数;逍遥散30g/kg、15g/kg剂量给药第1、2、3周可显著增加模型大鼠的糖水消耗量;30g/kg剂量对模型大鼠的学习记忆能力有一定改善趋势。各剂量组对海马和皮质中5-HT和DA含量的异常有不同程度的改善作用。结论:嗅球摘除抑郁大鼠模型建立成功。逍遥散对该抑郁模型动物的行为学及脑内单胺类神经递质的异常具有一定改善作用,表现出抗抑郁效应。Objective:To build the model of olfactory bulbectomezed(OB) depression rats and study the effects of xiaoyao powder to the rats of this model on the abnormal behavior and the monoamine neurotransmitter contents of brain.Methods:This OB depression model was produced by surgery.The experimental animal were randomly divided into normal control group,sham-operated control group,model control group,the dosages of 30g/kg、15g/kg、7.5g/kg xiaoyao powder groups,and the dosage of 10mg/kg amitriptyline group.Drugs were supplied for 4 weeks.The weights,the open-field behavior and the consumption of a sweet solution were measured.The learning and memory ability of rats was evaluated by Morris maze.The dopamine and serotonin contents in hippocampus and cortex of the brain were detected by ELISA.Results:After olfactory bulbectomized,the index of the model group contrast to the index of the normal group,the weight was decreased(P0.05),in the open-field test,the numbers of ambulations(line crossings with all four paws) were increased(P0.05),the numbers of rearings(raising two paws above the floor) were also increased(P0.05),the consumption of a sweet solution was decreased(P0.05).In the Morris maze,the latent period to find the plant was increased(P0.01),and the times crossing the plant area were decreased(P0.05),and the 5-HT contents of the hippocampus were decreased(P0.05).After drugs treatment for 4 weeks,some abnormal behaviors and the monoamine neurotransmitter contents of brain had been improved.Conclusion:The produce of OB depression rats was succeed,xiaoyao powder had a good influence on this model and displayed anti-depression effects.
