扎冲十三味及其有效组分挥发油对大鼠脑缺血的保护作用

摘　　要：目的:观察"扎冲十三味"及其有效组分挥发油对大鼠脑缺血的保护作用并探讨其作用机理。方法:将SD大鼠随即分为假手术组、模型组、扎冲十三味全方组(0.24g/kg)、扎冲十三味挥发油大剂量组(0.24g/kg)、扎冲十三味挥发油小剂量组(0.048g/kg)。各组灌胃给药27天后,采用线栓法制作MCAO脑缺血模型,测定大鼠神经功能评分、脑梗死指数以及大鼠血清炎症因子的表达。结果:与模型组比较,扎冲十三味全方组神经功能评分、脑梗死指数及血清IL-1、IL-6与MCP-1均显著降低(p<0.05);扎冲十三味挥发油大剂量组对局灶性脑缺血大鼠神经功能评分、梗死指数无明显影响,但血清炎症因子水平显著降低(p<0.05);扎冲十三味挥发油小剂量组对局灶性脑缺血大鼠神经功能评分、梗死指数及血清炎症因子水平均无明显影响(p>0.05)。结论:扎冲十三味抑制IL-1、IL-6和MCP-1等炎性因子的表达是其发挥保护缺血性脑损伤作用的重要机制之一,其中的挥发油可能通过抑制血清炎症因子的表达、与其他有效组分共同作用才能达到保护脑缺血损伤的目的。Objective：To observation the protective effects of ＂Garidi-13＂ and its effective fraction-the volatile oil on rats with cerebral ischemia.Methods：The rats were randomly divided into sham operation group,model group,Garidi-13 group（0.24g/kg）,volatile oil of large dose group（0.24g/kg）,volatile oil of small dose group（0.048g/kg）.After continuous intragenic administration 27ds,Rat model of focal cerebral ischemia was established by suture-occluded method,Neural function score and cerebral infarction index and expression of inflammatory factors in serum of rats were determined.Result：Compared with model group,the neurological function score and infarction index and levels of serum inflammatory factors of Garidi-13 group decreased significantly（p0.05）;The levels of serum inflammatory factors of high dose group volatile oil decreased differences（p0.05）;Volatile oil of small dose group have no difference index（p0.05）.Conclusion：Garidi-13 has protective effect on rats with cerebral ischemia.Adequate application of volatile oil can inhibite inflammatory factor.It is revealed that Garidi13 protection mechanism of action for cerebral ischemia may be associated with the drug combination,anti-inflammatory factor.
