摘  要:目的探讨金黄色葡萄球菌PV-杀白细胞素(PVL)对THP-1巨噬细胞活性及凋亡/坏死的作用。方法采用CCK-8比色法检测重组PV-杀白细胞素(rPVL)对THP-1巨噬细胞活性的影响;应用流式细胞仪检测THP-1巨噬细胞凋亡/坏死率;用Western blot法检测rPVL刺激THP-1巨噬细胞NF-κB的蛋白表达。结果随着rPVL对THP-1巨噬细胞作用的浓度和时间的增加,细胞活性进行性下降,在低浓度(5 nmol/L rPVL)以凋亡为主,在高浓度(40 nmol/L rPVL)以坏死为主,且具有时间依赖性。NF-κB的蛋白在高浓度(40 nmol/L rPVL)刺激下活化,2 h量表达最大,具有时间依赖性。结论 rPVL对THP-1细胞有毒性作用,低浓度诱导其凋亡、高浓度促进坏死,并能活化NF-κB的蛋白。Objective To investigate the effects of Panton-Valentine leucocidin (PVL) on cell viability of THP-1 macrophages and induction of its apoptosis/necrosis. Methods The cell viability of THP-1 macrophages induced by recombination Panton-Valentine ]eukocidin (rPVL) was determined by CCK-8 assay. The rates of cells apopto- sis/necrosis of THP-1 macrophages induced by different time and concentration of rPVL were detected by flow cy- tometry. Western blot method was used to test the expression of NF-KB protein in THP-1 macrophages induced by rPVL. Results After treatment with rPVL, the cell vitality decreased in time and concentration dependent man- ners. 5 nmol/L rPVL treatment group had higher apoptosis rate than that of control group and 40 nmol/L rPVL treatment group had higher necrosis rate than that of control group. And both of them were in the time-dependent manner. Western blot method showed that rPVL was able to induce expression of NF-KB in THP-1 macrophages in- duced by 40 nmol/L rPVL. Conclusion Low dose PVL induces apoptosis of THP-1 macrophages, and high dose rPVL directly induces necrosis. PVL can activate the expression of NF-KB protein.
