摘 要:为阐明泛素(Ubiquitin)在高等植物中结构与功能的关系,从千里光全长cDNA文库中分离到泛素基因,采用生物信息学方法对其进行系统分析。碱基序列和系统进化树结果显示,千里光泛素基因与果子蔓泛素基因的核苷酸序列(GenBank登录号:GQ890686.1)的同源性最高(87%);尽管碱基位点出现较大的变异,但高度保守氨基酸序列结果提示,泛素的保守位点对维持蛋白质结构和功能发挥关键作用;蛋白质的理化性质、三维结构预测与亚细胞定位结果表明,作为辅酶,泛素主要参与高等植物细胞的信号转导、转录调控和物质转运等功能。In the present study,the relationship of structure and functional role in ubiquitin is explored in Chinese medical plant such as Senecio scandens Buch.-Ham.ex D.Don,based on full-length cDNA library.Results of sequence analysis and phylogenetic tree of ubiquitin gene indicate that higher homology of ubiquitin gene is observed among Senecio scandens Buch.-Ham.ex D.Don.Moreover,the highest homology is detected in Guzmania lingulata(GenBank accession No.GQ890686.1)and Catharanthus roseus,with the nucleotide similarity(87%)and the amino acid sequence homology(97%).Those results indicate that the conserved amino acid mutations may play crucial roles in performing function of ubiquitin protein.Moreover,we also find that ubiquitin is mainly localized in the cytoplasm and mitochondrial matrix space in high plants.Accordingly,these results possibly suggested that ubiquitin could play an important role in signal transduction,transcription and transporter in the high plants,associated with its structural and functional predictions.
