烟草激活标签突变体库的创制和突变表型的初步鉴定

摘　　要：突变体库在烟草功能基因组研究中具有重要的作用,本研究建立了快速、高效农杆菌介导的烟草遗传转化体系,目前已经创制了3万份激活标签(pSK1015)突变体。利用PCR扩增对突变体库进行阳性检测,扩增结果表明其阳性率接近90%。大田T1代可视突变表型调查结果表明,在团棵期具有可视突变表型的概率为5.4%,在现蕾期具有可视突变表型的概率为3.6%。Mutants play important roles in tobacco functional genomic research.We established a rapid and efficient Agrobacterium tumefaciens mediated tobacco transformation protocol and have generated approximately 30 000 independent activation tagging lines with pSK1015 that possessed resistance to the herbicide glufosinate.PCR analysis showed that about 90% Basta resistant plant were transgene-positive plant.Field phenotyping of part of the activation tagging populations in T1 generations showed that approximately 5.4% and 3.6% of activation-tagged lines showed visible phenotypes at resettling stage and squaring stage,respectively.
