塞来昔布 真武汤逆转大鼠心肌肥厚的病理学对比

摘　　要：目的比较真武汤冲剂和塞来昔布胶囊逆转大鼠慢性压力负荷性心肌肥厚病理学的变化。方法将SD大鼠随机分为4组，假手术组8只，手术组、西药组和中药组各15只。手术组、西药组和中药组行腹主动脉缩窄术，制作心肌肥厚模型。16周行心脏超声，确定术后心肌肥厚，西药组加用塞来昔布胶囊混合饲料喂养，中药组加用真武汤混合饲料喂养，24周手术开胸，心脏组织进行病理学检查，包括HE染色、MASSION染色。结果HE染色中，手术组心肌细胞不同程度增大、核深染，纤维组织增生明显；中药组和西药组在不同程度上有所缓解。MASSION染色中，手术组胶原纤维增生明显，以血管周围明显；中药组和西药组胶原纤维增生较少，与手术组比较，血管周围减少更为明显。结论真武汤冲剂和塞来昔布胶囊均可逆转大鼠压力负荷性心肌肥厚，使心肌细胞增大、核深染、胶原纤维增生的程度减少。Objective To investigate the pathological changes in myocardial hypertrophy reversed by Zhenwu Tang granules and Celecoxib capsules in the rats with chronic pressure - overloaded left ventricular （LV） hypertrophy. Methods The experiment was performed in the laboratory of Guangzhou Institute of Respiratory Disease. All the male SD rats were randomized into 4 groups ： sham operation group（ n = 8 ）, operation group （ n = 15 ）, medicine group （ n = 15 ） and herbal medicine group （ n = 15 ）. The models of myocardial hypertrophy were made by gradually constricting the abdominal aorta. 16 weeks later, cardiac ultrasonography was performed in all the groups in order to ascertain post - operational left ventricular hypertrophy. And Zhenwu Tang granules were added in the herbal medicine group with the dose of 12 g/kg in the mixed feedstuff for 8 weeks,at the same time Celecoxib capsules were added in the medicine group with the dose of 10 mg/kg. In 24th week, we opened the chest by operation and took the cardiac muscle marked by HE and MASSION dyeing. Results The myocardial cells enlarged, nucleus color deepened and fiber tissue was apparent hyperplasia to different extent by HE dyeing in operation group, which was eased in some degree in the herbal medicine group and medicine group. Collagen fiber was apparent hyperplasia, particularly around the vessel by MASSION in operation group, but which was reduced in the herbal medicine group and medicine group. Hyperplasia was a little bit in herbal medicine group and medicine group, which was more than in sham operation group, but less than in operation group. Condasion Zhenwu Tang granule and Celecoxib capsule could reverse myocardial hypertrophy in rats with pressure overloaded left ventricular hypertrophy, and in this proeessmyocardial cells enlarge, nucleus color deepens and the hyperplasia of collagenous fiber decreases.
