心肌特殊染色在大鼠电击死中的应用

摘　　要：目的研究电击死大鼠的心脏在HE染色和Alcian blue-核固红染色下的形态学变化,为电击死和电损伤的鉴定提供一定依据。方法自行设计电击系统一套,雄性Wistar大鼠12只,分为正常对照组和电击死组,每组各6只。电击死后取大鼠心脏中性甲醛固定,制成切片后分别进行HE染色和Alcian blue-核固红染色。结果与正常组大鼠心肌染色相比,电击死组光镜下HE染色仅可见心肌纤维明显断裂,间质水肿、出血,而Alcian blue-核固红染色法可见心房和房间隔的特定区域组织蓝染。结论电击前后心脏HE染色与Alcian blue染色差异对电流损伤的研究有一定的参考价值。Objective To explore the morphological changes of myocardium after HE staining and Alcian bule staining,and to provide an evidence for identifying the electrocution from the electrical injury. Methods Twelve rats weighing 190 -210 g were divided into normal control group （n = 6） and electric shock group （n = 6）. After the rats were put to death,the samples were placed in 10% neu- tral buffered formalin and paraffined for HE staining and Alcian blue staining. Results Compared with controls,there was only a few fragmentation of myocardial fibrils around small vascular, interstitial bleeding and edema in electrocuted rats by HE staining. However, the blue staining of the myocardial fiber in electric shock group was observed by Alcian blue staining, and the results were obviously dif-ferent from normal group. Conclusion The difference of HE staining and Alcian bule staining for myocardium has a value to identify the electrogalvanic lesions.
