摘  要:目的对大黄中多糖成分进行提取、纯化,并对其组分及结构进行分析。方法采用超声提取大黄多糖,三氯乙酸脱蛋白,Superdex 200凝胶色谱纯化多糖,采用气相色谱分析多糖的组分,采用紫外光谱分析多糖结构。结果经分离纯化,得到单糖纯品SRPⅠ、SRPⅡ、SRPⅢ,其中SRPⅠ由葡萄糖、半乳糖和木糖组成,SRPⅡ由鼠李糖、葡萄糖、阿拉伯糖和半乳糖组成,SRPⅢ由鼠李糖、半乳糖、岩藻糖、甘露糖及果糖组成,紫外光谱分析发现,大黄多糖中含有β型糖苷键、吡喃糖环、氢键及羟基等结构。结论大黄多糖含有多种单糖组分,分子结构较为复杂。Objective To extract and purify polysaccharide from rhubarb,and analyze to composition and construction of rhubarb polysaceharide. Methods Rhubarb polysaccharide were extracted with ultrasound extraction ways, and were de- proteinized with trichloroacetic acid,were purified with Superdex 200 gel chromatography, composition of rhubarb polysac- eharide were analyzed with gas chromatogram ways, construction of rhubarb polysaccharide were analyzed with ultraviolet spectra. Results Pure monosaccharide that was SRP Ⅰ, SRP Ⅱ and SRP m were were obtained by segregation and purifica- tion, SRⅠ included glucose, galactose and xylose, SRPⅢ included rhamnose, glucose , arabinose and galactose, SRP Ⅲincluded rhamnose, galaetose, fucose, mannose and fructose. Ultraviolet spectra shown construction of rhubarb polysaccha- ride include β-glycosidic linkage, pyran ring, hydrogen bond and hydroxyl. Conclusion Construction of rhubarb polysac- charide included many kinds of monosaccharide and construction was complicating.
