喂饲转HJC-1和G6-EPSPS基因抗虫耐草甘膦大米大鼠亚慢性毒性试验

摘　　要：目的研究喂饲转HJC-1和G6-EPSPS基因抗虫耐草甘膦大米对大鼠的亚慢性毒性。方法将140只健康6～8周龄SPF级Wistar大鼠按体重随机分成7组,分别为饲养对照组和低(17.5%)、中(35%)、高(70%)剂量转基因大米组及低(17.5%)、中(35%)、高(70%)剂量亲本组,每组20只,雌雄各半。采用饲喂方式染毒,连续饲养90 d。观察大鼠的一般情况,测定体重、进食量;在实验中期(第45天)和实验结束时(第90天)检测血液学和血生化指标,在实验结束后测定各主要脏器重量,计算脏器系数;并进行脏器病理组织学检查。结果各剂量组动物的体重均呈增长趋势,转基因组、亲本对照组与饲养对照组每周体重及体重总增重、每周进食量、总食物利用率以及中、末期的血液学和血生化指标,各主要脏器系数及病理组织学检查比较,均未发现有生物学意义的改变。结论未发现喂饲转HJC-1和G6-EPSPS基因抗虫耐草甘膦大米对大鼠具有亚慢性毒性。Objective To observe the subchronic toxic effects of transgenic rice on rats. Methods Based on gender and weight, 140 Wistar rats were randomly divided into seven groups with 20 rats each group： three transgenie rice groups with different doses, three non-transgenic rice groups with different doses and one normal control group. The diets of transgenie rice groups contain 17.5%, 35% and 70% transgenic rice respectively. The diets of non-transgenic rice groups contained 17.5%, 35% and 70% non-transgenic rice respectively. The diet of control group contained only basic feed.They were fed for 90 consecutive days. The blood routine test,blood biochemistry test （at the 45 th day and the 90 th day of the experiment）, organ weight and organ pathological examination （at the end of the 90 th day） were conducted. Results No transgenic HJC-1 and G6-EPSPS rice related adverse effects on the body weight, food intake, food consumption, hematology, serum biochemistry, as well as histopathology were seen. Conclusion In the present study, there are no enough evidences to confirm that transgenie HJC-1 and G6-EPSPS rice has subchronic toxic effects on rat.
