摘  要:目的研究nodosin对大鼠胶质瘤C6细胞的抗肿瘤作用。方法采用倒置显微镜对C6细胞进行形态学观察;采用四甲基偶氮唑蓝(MTT)法测定细胞生存率,乳酸脱氢酶(LDH)法检测细胞毒性,BrdU检测法检测细胞增殖情况;采用Hoechst33342染色法观察nodosin对C6细胞凋亡的影响。结果Nodosin在作用12h和24h可抑制大鼠胶质瘤c6细胞的细胞生存率,呈时间与剂量依赖性;在0~20μmol·L。浓度范围内对C6细胞无细胞毒性作用;20μmol·L—nodosin作用24h,C6细胞增殖率降低至74.6%;同时Nodosin能够促进C6细胞的凋亡。结论Nodosin具有抗肿瘤作用,其作用与抑制细胞的增殖及促进细胞的凋亡有关。Objective To evaluate the antitumor effect of nodosin on rat C6 glioma cells. Methods Morphologic observation of nodosin on C6 cells was performed under phase contrast microscope, cell viability was tested by MTT assay, and cell proliferation was detected by BrdU assay. The cytotoxicity of nodosin was evaluated by lactate dehydrogenase (LDH) leakage method and Hoechst 33342 staining was used to test the cell apoptosis. Results Nodosin could inhibit the cell viability of the rat C6 glioma cells in time-dependent and dose-dependent manner at hour 12 and 24, and it had no cytotoxicity on C6 glioma cells within 20 μmol· L-1. The cell proliferation rate was reduced to 74.6 % after treatment with 20μmol·L-1 of nodosin at hour 24, and nodosin promoted the apoptosis of C6 cells. Conclusion Nodosin has antitumor action on C6 cells by inhibiting cell proliferation and inducing apoptosis.
