摘 要:目的探讨毛冬青总黄酮对大鼠脑缺血模型的作用特点及其机制。方法大鼠连续灌服不同剂量毛冬青总黄酮10 d,于第10天给药物后1 h,大鼠两侧颈总动脉用微动脉夹夹闭30 min,复制脑缺血模型;取大鼠脑,一半脑组织用生理盐水制脑匀浆,测脑匀浆三磷酸腺苷(ATP)酶水平,乳酸(LD)、丙二醛(MDA)含量;另一半脑组织固定、切片,观察脑组织的病理变化。结果微动脉夹夹闭双侧颈总动脉可成功复制脑缺血模型。与模型组比较,毛冬青总黄酮可显著提高脑匀浆Na+-K+-ATP、Mg2+-ATP、Ca2+-ATP和Ca2+-Mg2+-ATP酶活力(P<0.01,P<0.05),明显降低LD含量(P<0.05),显著减少MDA的含量(P<0.01),显著减轻缺血所致的脑组织损伤。结论毛冬青总黄酮对大鼠脑缺血模型有较好的改善作用。Objective To explore the effect of total flavonoids from Radix Ilecis Pubescens (Maodongqing) on rat cerebral ischemia models and to explore its mechanism. Methods Rats were fed with large-, middle- and small-dose suspension of total flavonoids from Radix Ilecis Pubescens for 10 continuous days. One hour after the administration on the 10th day, we blocked the bilateral common carotid arteries of rats for 30 min with arteriole clipping to induce cerebral ischemia, At the end of the experiment, adenosine triphosphate (ATP), lactic dehydrogenase (LD) and malondialdehyde (MDA) contents in the cerebral homogenate were examined. The pathological changes of brain tissue were also observed. Results Cerebral ischemia rats model was established successfully by occlusion of bilateral common carotid artery. Compared with the model group, the activities of Na+-K+-ATP, Mg2+-ATP, Ca2+-ATP and Ca2+-Mga+-ATP in brain homogenate were enhanced(P 〈 0.01, P 〈 0.05), LD content was significant decreased(P 〈 0.05), the production of MDA was significantly reduced(P 〈 0.01 ), and the ischemia-induced brain damage was significantly relieved in total flavonoids groups. Conclusion Total Flavonoids from Radix Ilecis Pubescens show satisfactory effect on improving cerebral ischemia function in rat models.
