摘 要:目的:探讨地榆总皂苷体内抗肿瘤血管生成作用。方法:构建小鼠移植性S180皮下瘤模型,观察地榆总皂苷对荷瘤小鼠肿瘤生长抑制作用;同时解剖瘤组织,采用免疫组化法(IHC)检测地榆总皂苷对小鼠肿瘤组织微血管VEGF表达水平,染色结果采用半定量分析。结果:地榆总皂苷灌胃给药剂量4g/kg、2g/kg、1g/kg对荷S180肉瘤小鼠肿瘤生长抑制率分别为38.65%、30.81%、24.72%,显微镜下观察,地榆总皂苷作用后各组小鼠肿瘤组织微血管数较阴性对照组明显减少,阴性对照组、地榆总皂苷高、中、低剂量组VEGF蛋白IHC染色综合评分分别为11.72±2.34、1.04±0.02、6.83±0.51、9.82±0.43,经统计分析,地榆总皂苷各剂量组IHC染色综合评分较阴性对照组低,差异均有显著性(P<0.05)。结论:地榆总皂苷具有一定的体内抗肿瘤血管生成作用,其作用与抑制肿瘤组织VEGF表达水平有关。Objective : To study the antiangiogenesis effect of Sanguisorba saponins in vivo. Methods : Mouse S180 - tumor models were established and divided randomly into five groups as follows: negative control group (0.5 % CMC) , positive control group ( FT - 207,120 mg/kg), Sanguisorba saponins high dose group ( 4g/kg), medium dose group ( 2g/kg), low dose group (lg/kg). After intragastric administrated for 10 days continuously, all mice were sacrificed then the tumors were excised and weighed, then fixed in 10% neutral buffered formlin overnight, the growth inhibition was calcu- lated. IHC was performed to evaluate the expression of vascular epithelial growth factor (VEGF) in the toumors. Results: With 4g/kg, 2g/kg , lg/kg intragastric administration, Sanguisorba saponins could inhibit S180 by 38.65% , 30. 81% ,24.72% respectively. The amount of microvessels in each Sanguisorba saponins group was lower than that in the negative control group. By semiquantitative analysis, the staining scores of negative control group, positive control group Sanguisorba saponins high dose group, medium dose group, low dose group were 11.72 ± 2.34,1.04 ± 0.02, 6. 83 ±0.51,9.82 ±0.43 respectively. After statistical analysis,each staining scores of Sanguisorba saponins group was lower than those in the negative control group and the difference was significant(P 〈 0.05 ). It is proved that Sanguisor- ba saponins could both inhibit the growth of tumor microvessels and expression of VEGF in mice efficiently. Conclusion : Sanguisorba saponins has an antiangiogenesis effect in vivo, which is related to the inhibition of the expression of VEGF in tumors.
