高含油量甘蓝型油菜小孢子培养体系探究

摘　　要：以8个高含油量甘蓝型油菜品系的杂交F1为试验材料,观察小孢子各发育时期的形态,探讨花蕾长度与小孢子发育时期对应关系,培养基中添加不同植物激素及质量浓度配比等因素对小孢子培养出胚量及再生胚生长的影响。结果表明,小孢子不同发育时期的形状差异较大,小孢子单核晚期对应的花蕾长度主要集中在2.5～3.0mm;只有少数膨大细胞最终发育成为胚状体;不同材料出胚量差异较大,有5个材料获得再生胚,其中2003×HZ出胚量最高,为每蕾8.1胚,另3个材料未获得再生胚;不同材料小孢子对激素种类和质量浓度的反应不同,在NLN中添加0.1mg/L 2,4-D+0.05mg/L(或0.1mg/L)6-BA利于2003×HZ、1517×HZ、1485×HZ出胚,而在NLN中添加0.05mg/L NAA+0.05mg/L 6-BA利于1438×HZ、1479×HZ出胚;在B5固体培养基中添加0.1mg/L 2,4-D+0.1mg/L 6-BA利于胚状体迅速生长。In order to establish a microspore cultivation system for eight F1 hybrids of Brassica napus of high oil content, we observed the forms of microspores in each development stage, and explored the correspondence between the bud length and the microspores development stage. The effect on the out- puts of microspore and the embryos regenerating by various pbytohormone, diverse concentration rati- o and other factors were investigated. We found that the difference among various microspore develop- ment stages were remarkable. Flower bud length corresponded to late microspore mononuclear stage was 2.5 to 3.0 mm. Only a few of expanding cells could develop into embryoid. The outputs with va- rious materials were obviously different, three of the eight materials had no embryos, and 2003N HZ possessed 8.1 embryos per bud, which is the highest level of outputs of the rest five materials with embryos. Different materials reacted differently to hormones, NLN added with 0. 1 mg/L 2,4-D -k 0.05 mg/L （or 0.1 mg/L） 6-BA was beneficial to the embryos producing from 2003 N HZ, 1517 N HZ and 1485N HZ. NLN added with 0.05 mg/L NAA ＋ 0.05 mg/L 6-BA was in favor of the embryos generation from 1438N HZ and 1479 X HZ. Bs added with 0.1 mg/L 2,4-D ＋ 0.1 mg/L 6-BA availed the growth of embryoid.
