摘  要:利用RT-PCR和RACE法从新疆早实核桃新早丰花芽中克隆到控制植物成花的关键基因LEAFY,其cDNA全长为1 472bp(GenBank登录号为JF520778),含有1 155bp的开放阅读框,编码385个氨基酸。序列分析表明,该基因含有3个外显子和2个内含子,与其他植物上相关同源基因的结构相似,且与已克隆的LEAFY序列同源性较高。部分序列在早实和晚实核桃上进行扩增发现,该基因在早实核桃及晚实核桃上都能够得到扩增产物,且扩增产物的亮度早实核桃大于晚实核桃。在早实核桃不同组织器官之间进行扩增发现,该基因在早实核桃营养器官上的表达方式显示出自身的特有表达特征。The LEAFY homologous gene was cloned from the flower-bud of precocious walnut "Xinza- ofeng" in Xinjiang province by RT-PCR and RACE. The full length of cDNA was 1 472 bp(GenBank accession number:JF520778), and has a 1 155 bp open reading frame which encoding 385 amino acids. It shared high similarity with LEAFY homologues both at nucleotide acid and amino acid levels. There were three exons and two introns in the DNA sequence; it was similar to other plant LEAFY homolo- gous genes. The amplified product could be obtained both in precocious walnut and later-seeding wal- nut, but the brightness of amplification in precocious walnut was higher than that in later-seeding walnut. The expression pattern among different tissues of precocious walnut showed its unique char- acterlstlcs.
