摘  要:目的 研究紫外线 (UVB)诱导成纤维细胞 (NIH3T3 )凋亡时胞浆pH值的变化。方法 利用粘附式细胞仪(ACAS -5 70 )测定UVB诱导NIH3T3细胞凋亡时 ,胞浆pH值的变化。结果 NIH3T3细胞经UVB照射后 ,胞内pH值会逐渐升高 ,并在 1 6h左右达到最大值 ,2 5min后恢复正常 ,在细胞内外钙被螯合后 ,不管UVB照射与否 ,胞内pH值无变化。结论 UVB照射导致细胞凋亡时 ,胞液呈碱性 ,pH值的变化参与了细胞凋亡的调控 ;细胞内外Ca2 + 水平影响胞浆pH值的变化 ,胞浆Ca2 + 与胞浆pH值的变化存在一定的相关关系。Objective To explore the changes of intracellular pH in the apoptotic fibroblast (NIH3T3 cells) induced by UVB radiation. Methods\ By using adherent cell analysis and sorting (ACAS-570), the changes of intracellular pH were determinated while apoptosis was induced in NIH3T3 cells by UVB.Results\ The intracellular pH of the NIH3T3 cells radiated by UVB rose and reached the highest level after 1h and 35mins then returned to normal in 25mins. When extracellular and intracellular calcium were chelated,the intracellular pH of the NIH3T3 cell, radiated by UVB or not, had no changes. Conclusion\ The intracellular pH rises in the apoptotic NIH3T3 cells induced by UVB radiation. The changes of intracellular pH, affected by cytosol Ca 2+ level, participate in the regulation of apoptosis. Regulation of apoptosis and were affected by the level of cytosol Ca 2+ . There is correlation between cytosol Ca 2+ and intracellular pH.
