牛脑中性鞘糖脂Glob-N的分离鉴定

摘　　要：为了探索一种新的中性鞘糖脂 (neutral glycosphingolipids,N- GSL s)单一组分的纯化分离鉴定方法 ,将通过改良的 Hakomori方法提取的牛脑中性鞘糖脂经过加压柱层析 ,梯度甲醇 /氯仿溶液洗脱。所得到的产物行 1 H- NMR(核磁共振 )和 1 3C- NMR测定。结果发现含甲醇 6 0 %的甲醇 /氯仿溶液的洗脱物为纯化 Glob- N (红细胞糖苷脂 ) ,层析展开后呈单一清晰条带 ,Rf值 0 .45。初步结构分析表明牛脑 Glob- N含 N-乙酰半乳糖 ,共 3个糖基 ,其神经酰胺部分含有 2 6个碳。结果表明柱层析分离中性鞘糖脂单一组分效果好 ,纯度高。牛脑 Glob- N结构中含In order to establish a new method of isolating individual component of neutral glycosphingolipids (N GSLs), N GSLs from cow brain was studied. Glob N from cow brain was separated by column chromatography with methanol/chloroform solution for gradient elution, and its structure was identified by 1H NMR and 13 C NMR. The results showed that Glob could be obtained by elution with 60 percent of methanol in methanol/chloroform solution(by vol), with the Rf value 0.45. The structural analysis showed that Glob N from cow brain was a kind of globotriao ceramide, containing GalNAc with 26 C in its ceramide moiety. Our results suggested that individual N GSL could be well separated from total N GSLs by column chromatography with high production. The Glob N from cow brain was a kind of globotriao syl ceramide.
