摘 要:目的 :研究不同膳食组成对小鼠肌肉胰岛素受体及其相关底物基因表达的影响。 方法 :C5 7BL / 6 J小鼠分别饲以高脂饮食、高脂加谷氨酰胺饮食、高脂 3个月后补加谷氨酰胺饮食以及正常对照饮食 ,实验期为 5 .5个月。以反转录 -聚合酶链反应 (RT- PCR)分别测定肌肉胰岛素受体 (IR)及其受体底物 1(IRS- 1)、受体底物 2 (IRS- 2 )以及磷酸肌醇 3激酶 (PI- 3- kinase)m RNA水平。 结果 :高脂可使小鼠肌肉 IR,IRS- 1,IRS- 2 ,PI- 3- kinase m RNA水平不同程度下降 ,分别为正常对照的 81% ,6 3% ,5 9%及 45 %。结论 :谷氨酰胺可在一定程度上抑制上述基因 m RNA下降的趋势 ,提示营养因素对机体胰岛素敏感性的影响可能部分是通过影响其胰岛素受体及其一系列相关底物的基因表达而实现的。Objective: To investigate the influence of nutrient on the expression of insulin receptor and its related substrate genes in the muscle of mice. Methods:The C57BL/6J mice were divided into 4 groups randomly :(1)Fat,mice on high fat diet;(2)F+G,supplemented with glutamine after 3 month on high fat diet;(3)Gln,high fat with glutamine; (4)Control, normal diet.Each group was fed for 5.5 month. Reverse transcription polymerase chain reaction(RT PCR) was used to determine the mRNA levels of insulin receptor (IR),insulin receptor substrate 1(IRS 1),insulin receptor substrate 2(IRS 2) and phosphatidylinositol 3 kinase(PI 3 kinase).Results: mRNA level of IR,IRS 1,IRS 2,and PI 3 kinase in the mice on high fat diet were lowered to some degree: 81%,63%,59% and 45% that of control. Conclusion: Glutamine can prevent the reduction of the mRNA level of the genes mentioned above in fat group to some extent.The results suggest that nutrient may influence insulin sensitivity of the body through regulation of genes mentioned above.
