摘  要:目的建立实验秃毛大鼠模型,研究重组人角质细胞生长因子-2(recombinant human keratinocyte growth factor-2,rhKGF-2)对实验秃毛大鼠的毛发再生作用。方法对大鼠背部以脱毛膏拔毛,随机分为7个组,即正常对照组、5%米诺地尔组、aFGF组、rhKGF2高、中、低剂量组、0.9%生理盐水组。造模次日皮肤涂抹给药,以给药第1、5、9、14天为观察时间点,记录实验大鼠的体重、实验区毛长、实验区毛发生长状况。于第14天处死实验大鼠,取实验区皮肤进行组织病理学检查。结果体重方面:在第1、5、9、14天,与正常对照组相比,各组差异无显著性(P>0.05);毛长方面:与0.9%生理盐水组相比,在第5、9天,rhKGF2高、中剂量组存在显著性差异(P<0.05,P<0.01)。在第14天,高、中、低剂量组差异有显著性(P<0.05,P<0.01,P<0.05);毛重方面:在第14天,与0.9%生理盐水组相比,各组(正常对照组除外)差异有显著性(P<0.01);病理检查结果显示:rhKGF2具有促进实验秃毛大鼠毛囊新生与成熟的作用。结论 rh-KGF2具有促进实验秃毛大鼠毛发再生的作用。rhKGF2具有很好的临床应用前景。Aim To establish the experimental bald rat model and study the effect of recombinant human keratinocyte growth factor-2(rhKGF2)on hair regeneration in experimental bald rat.Methods The hair of SD rats was removed by paste,and these SD rats were then randomly divided into seven groups,including normal group,5% minoxidil group,aFGF group,rhKGF2 high-dose group,rhKGF2 middle-dose group,rhKGF2 low-dose group,0.9% saline group.Skin was treated with different medications as described above the day after hair removal.Body weight,hair length,hair weight,and the hair growth conditions were recorded on days 1,5,9 and 14 after treatment.The rats were sacrificed on day 14,and the experimental skin was used for histopathological examination.Results Body weight of rats: compared with normal group,there was no significant difference in each group(P0.05) on days 1,5,9,and 14;Hair length of rats: compared to 0.9% saline group,there were significant differences in rhKGF2 high-dose(P0.05) and rhKGF2 middle-dose groups(P0.01) on days 5 and 9.On day 14,compared with 0.9% saline group,there were significant differences in rhKGF2 high(P0.05),middle(P0.01) and low-dose(P0.05) groups;Hair weight of rats: On day 14,compared with 0.9% saline group,there were significant differences among ather groups(except for normal group)(P0.01);Histological findings: RhKGF2 played an important role in the formation and maturation of hair follicles in the experimental bald rats.Conclusion RhKGF2 plays an important role in hair regeneration in the experimental bald rats.RhKGF2 has good prospects for clinical application.
