摘　　要：目的　研究gp130单克隆抗体B S12的生物学特性。方法　用细胞计数和3 H TdR掺入的方法观察人多发性骨髓瘤细胞株XG 1和XG 2在抗gp130的单抗B S12与它们的Fab片段和F(ab′) 2 片段作用下 ,细胞增殖和生长情况 ;此外 ,也观察gp130单抗B P8以及IL 6和IL 6R单抗B E8和B R6对B S12单抗在这 2个细胞株上作用的影响。结果　B S12可刺激XG 1和XG 2细胞增殖 ,并维持它们的长期生长 ,但B S12的这些作用有赖于它保持完整的二价结合力。B E8和B R6对B S12的作用无影响 ,B P8则可协同增强B S12的作用。结论　B S12为刺激型抗gp130的单抗 ,可起着类似于IL 6的作用。Objective To study the biological characteristics of gp130 monoclonal antibody (McAb) B S12. Methods Cell count and 3H TdR incorporation were used to assess the cellular proliferation and growth of human multiple myeloma cell lines XG 1 and XG 2 stimulated by both B S12 and the Fab and F(ab′) 2 fragments. In addition, the influences of anther gp130 McAb B P8, IL 6 McAb B E8 and IL 6R McAb B R6 on the effects of B S12 on the proliferation of XG 1 and XG 2 cells were observed. Results B S12 stimulated proliferation of XG 1 and XG 2 cells, and supported their long term growth. But, the effect of B S12 was dependent on keeping its intact binding ability. B E8 and B R6 had no influence on the effect of B S12, while B P8 could synergize and augment its effect. Conclusion B S12 is a stimulating gp130 McAb, and play a similar role to that of IL 6.
