不同组方祛白脂对小鼠B16黑素瘤细胞代谢的影响

摘  要:目的:探寻祛白脂组方中影响小鼠B16黑素瘤细胞代谢的因素。方法:以小鼠B16细胞增殖活性、黑素生成量、酪氨酸酶活性为指标,以8-甲氧补骨脂素(8-MOP)浓度和辅药为考察因素,选用3×2析因设计研究不同组方祛白脂对小鼠B16黑素瘤细胞代谢的影响,用SPSSl 3.0软件辅助数据分析。结果:8-MOP 10μmol·L-1处理组小鼠B16黑素瘤细胞增殖能力明显高于其他组(P<0.01),8-MOP 50μmol·L-1处理组小鼠B16黑素瘤细胞黑素含量、酪氨酸酶活性明显高于其他组(P<0.01);加入辅药小鼠B16黑素瘤细胞增殖能力、黑素含量、酪氨酸酶活性显著高于不加辅药组(P<0.01);8-MOP浓度和辅药处理的交互作用对小鼠B16黑素瘤细胞增殖能力、黑素含量、酪氨酸酶活性有显著影响(P<0.01)。结论:祛白脂中8-MOP浓度和辅药是影响小鼠B16黑素瘤细胞代谢的主要因素。Objective: To explore the impact factors in the formula of Qubai creams on the metabolism of murine B16 melanoma cells.Method: The 3 ×2 factorial design was used in the different formula of Qubai creams with the 8-MOP concentration and supplementary drug as the independent variables.Proliferation,melonogenesis and tyrosinase activity of the cultured B16 melanoma cells affected by the creams were determined and analyzed by SPSSl3.0.Result: The proliferation of the cultured B16 melanoma cells affected by 10 μmol·L-1 8-MOP was significantly higher than that of the others,the melonogenesis and tyrosinase activity in the cultured B16 melanoma cells affected by 50 μmol·L-1 8-MOP were significantly higher than those of the others.The proliferation,melonogenesis and tyrosinase activity of the cultured B16 melanoma cells affected by the supplementary drug was significantly higher than those without the supplementary drug(P0.01).The concentration of 8-MOP and the supplementary drug had remarkable interaction in the proliferation,melonogenesis and tyrosinase activity of the cultured B16 melanoma cells(P0.01).Conclusion: The concentration of 8-MOP and the supplementary drug are the main impact factors of Qubai creams in the metabolism of murine B16 melanoma cells.
