摘　　要：目的研究ICOS—GL50信号在T细胞依赖性B细胞体外增殖、抗体分泌中的作用，进一步探讨细胞因子IL-10对ICOS-GL50信号介导的体液免疫中的作用。方法分别采用台盼蓝活细胞计数法和ELISA法检测ICOS—L929基因转染细胞和细胞因子IL—10对PWM介导的B细胞体外增殖及分泌抗体的效应。结果ICOS-L929基因转染细胞能够促进PWM介导的B细胞体外增殖，而不能显著上调抗体的分泌；加入IL-10后，能显著促进PWM介导的B细胞体外增殖和抗体分泌。结论细胞因子IL-10在ICOS—GL50信号介导的机体T细胞依赖的体液免疫应答反应中发挥着重要的协同作用。Objective To understand the role of ICOS-GL50 signal in T-cell dependent B cells antibody production in vitro and further investigate the role of IL-10 in ICOS-GL50 mediated humoral immune responses. Methods ICOS-L929 transfectants and cytokine IL-10 were employed to study their impact on the PWM driven B cells proliferation and antibodies secretion by Trypan blue staining and ELISA assay.Results ICOS-L929 transfectants could promote the proliferation of PWM-driven B cells but not increase the antibody production. However,the level of antibody secretion from PWM-driven B cells was up-regulated in presence of IL-10. Conclusion IL-10 plays an important synergetic effect on ICOS-GL50 signal mediated secondary T cell dependent humoral immune responses.
