摘 要:目前研究者大都采用愈伤组织诱导法或水培法进行马铃薯抗性的早期评价,受自然因素影响较大。本试验以无菌试管苗和马铃薯晚疫病菌分泌物为实验材料,建立试管苗室内评价技术体系并通过分泌物接种试验探测其可行性,以便用于早期抗性资源的评价,缩短育种周期。本试验在无菌条件下进行,将健康试管苗植于含有马铃薯晚疫病菌株XH05-5-4分泌物的MS培养基中培养,培养1个月后,调查其生长情况。结果表明:马铃薯试管苗在接种分泌物培养1个月后,处理组试管苗明显矮于空白对照组试管苗。处理组试管苗叶片有褐变现象,同时根的分化受阻。不同品种(系)做同样处理后株高比空白对照组的株高减少的程度不同,可得知不同品种(系)的马铃薯试管苗的抗性不同。因此,用分泌物接种试管苗来进行抗性评价是可行的,有利于进行对抗性资源的早期评价,减少抗病育种的工作量,缩短育种周期。Researchers mostly adopt callus induction method or hydroponic method for potato early re- sistance evaluation. It's generally based on field trials by natural factors. In the test, we used the pla- ntlets of sterile room and the secretion of Phytophthora infestans as experimental materials. The labora- tory evaluation system of the virus-free plantlets was established and its feasibility was detected by se- cretion inoculation experiments, which was used for the early evaluation of resistance to the resources and shorten the breeding cycle. The test was carried out under sterile conditions. The healthy plantlets were implanted in MS medium containing secretion of Phy-tophthora infestans XH05-5-4. We would in- vestigate the growing situation a month later. The results indicated that after inoculation of secretion one month, the plantlets tested were significantly shorter than the plantlets of the vacuity contrast group. Also, the leaves of plantlets tested were brown and the differentiation of roots was blocked. The plant height of different varieties (lines) was decreased differently by the same test. It showed that the different varieties (lines) of potato plantlets had the different fastness. So the resistance evaluation of plantlets was feasible with the inoculation of secretion. It was used for the early evaluation of resistance to the resources, to reduce the workload of breeding and shorten the breeding cycle.
