摘　　要：目的　探讨CD40分子在树突状细胞和外周血单核细胞表达的意义。方法　将人外周血单核细胞经不同组合的细胞因子共培养获得了树突状细胞 ,经流式细胞仪表型分析、活细胞计数、混合淋巴细胞反应等方法测定抗人CD40单抗对CD40分子的激发作用。结果　激发型抗人CD40单克隆抗体 5C11能促使树突状细胞分化成熟 ,并且激发后的DCs 具有明显的激发T细胞的功能。结论　抗人CD40激发型单抗是一种潜在的免疫激动剂。Ojective To study the function of CD40 molecules expressed on peripheral monocytes and dendritic cells.Methods Dendritic cells were induced after peripheral monocytes had been cultured in medium with different combination of cytokines;Methods of flow cytomete analysis,cell counting,mixed lymphocyte reaction were used to study the function of agonistic anti-CD40 mAb 5C11.Results Agonistic anti-CD40 monoclonal antibody 5C11 induced formation and maturation of dendritic cells from peripheral monocytes and the DCs induced strongly stimulated T cell activation.Conclusion MAb 5C11 is a potential immunologic stimulator.
