摘 要:目的 :探讨银杏叶提取物 ( Egb76 1)在心肌缺血再灌注损伤中的预防作用。方法 :取 30只 SD大鼠 ,随机平均分为 3组 ,采用改良 L angendorff法建立离体大鼠心脏缺血再灌注模型 ,以 Krebs- Henseleit碳酸氢盐缓冲液 ( KHBB)灌注 10 min后 ,实验组和对照组分别予以 Egb76 1加晶体停搏液或晶体停搏液停搏 ,停搏 6 0 m in后再以 KHBB缓冲液灌注 6 0 m in,在复灌15、30、6 0 min时点测定冠脉流量、心率、左心室压力 ,并收集心脏灌流液测定乳酸脱氢酶 ( L DH )、肌酸磷酸激酶 ( CPK ) ;心肌标本行电镜检查。结果 :心肌复灌早期 ,实验组与对照组冠脉流量均较停搏前减少 ,但实验组恢复较快 ( P<0 .0 5 ) ,冠脉流量较对照组明显增大 ( P<0 .0 5 ) ,心率也较快恢复至停搏前水平 ;L DH与 CPK实验组明显低于对照组 ( P<0 .0 1,P<0 .0 0 1)。停搏后实验组与对照组冠脉流量均较停搏前减少 ,但实验组恢复较快 ,两者有显著性差异 ( P<0 .0 5 ) ;复灌后 ,实验组心率也较快恢复至停搏前水平 ;电镜观察显示实验组心肌超微结构改变明显小于对照组。结论 :在离体大鼠心肌缺血再灌注损伤过程中 ,Egb76AIM: To study the in vitro preventive effect of Ginkgo biloba extracts (Egb761) on myocardial ischemic reperfusive injury in SD rats. METHODS: Thirty SD rats, which were divided into 3 groups (n 1=n 2=n 3=10), were chosen to make the model of myocardial ischemic reperfusive injury of SD rats by using developed langendorff methods. After 10 min of perfusion with Krebs Henseleit bicarbonate buffer (KHBB), the hearts of SD rats in the test group were arrested by the cardioplegia with Egb761, while those in the placebo group were arrested only by the cardioplegia without Egb761. Sixty min later, all the hearts of SD rats were reperfused by the KHBB, and the coronary flow, heart rate, left ventricular pressure were measured at 15, 30, 60 min after reperfusion, the perfusive fluid of the hearts were collected to measure the value of lactate dehydrogenase (LDH) and creatine phosphokinase (CPK). The myocardia of the rats were tested by the electron microscope. RESULTS: The coronary flow decreased both in the test group and in the placebo group after the early reperfusion, but it recovered more quickly and more significantly in the test group (P<005). The values of LDH and CPK in the test group were lower than those in the placebo group (P<001,P<0001) and the changes of the myocardial ultramicrostructure were less significant in the test group. CONCLUSION: It is suggested that Egb761 could prevent the myocardial ischemic reperfusive injury in SD rats.
