摘 要:目的:观察葫芦巴总皂苷(trigonella foenumgraecum L.total saponin,TFGs)对高糖高脂饮食诱导的营养性肥胖大鼠的治疗作用.方法:采用高糖高脂饮食建立营养性肥胖大鼠模型,TFGs干预8wk.观察大鼠体质量,附睾周围脂肪组织质量,HE染色观察形态学.检测甘油三酯(triglyceride,TG)、总胆固醇(cholesterol,TC)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、空腹血糖(Fasting blood glucose,FBG)和空腹胰岛素(fasting insulin,FINS),计算胰岛素抵抗指数(homeostasis model assessment-insulin resistance,HOMA-IR).结果:与普通饮食大鼠相比,高糖高脂喂养肥胖大鼠体质量明显增加(495.5g±69.2gvs388.7g±60.2g,P<0.01),附睾周围脂肪显著增多(8.6g±0.6gvs5.3g±0.5g,P<0.01),脂肪细胞肥大,TG、TC和LDL-C水平明显升高(0.9mmol/L±0.3mmol/Lvs0.3mmol/L±0.2mmol/L,2.7mmol/L±0.4mmol/Lvs1.5mmol/L±0.1mmol/L,1.2mmol/L±0.2mmol/Lvs0.5mmol/L±0.2mmol/L,均P<0.01),HDL-C水平降低(0.5mmol/L±0.1mmol/Lvs0.7mmol/L±0.2mmol/L,P<0.01),FINS和HOMA-IR水平显著增高(27.8mIU/L±4.8mIU/Lvs13.6mIU/L±3.6mIU/L,8.0±1.1vs3.5±1.1,均P<0.01).与肥胖大鼠相比,TFGs治疗组大鼠体质量和附睾周围脂肪沉积显著减轻(462.5g±55.2gvs495.5g±69.2g,P<0.01;6.9g±0.3gvs8.6g±0.6g,P<0.01),脂肪细胞增生肥大受到抑制.同时,TG、TC、LDL-C和HOMA-IR水平显著降低(0.7mmol/L±0.2mmol/Lvs0.9mmol/L±0.3mmol/L,P<0.01;2.1mmol/L±0.3mmol/Lvs2.7mmol/L±0.4mmol/L,P<0.05;0.9mmol/L±0.1mmol/Lvs1.2mmol/L±0.2mmol/L,P<0.01;4.9±1.4vs8.0±1.1,P<0.01).结论:TFGs对营养性肥胖大鼠具有良好的糖脂调节作用,可能是治疗肥胖的有效药物.AIM: To observe the therapeutic effect of trigo- nella foenum-graecum L. total saponins (TFGs) on high-sugar and high-fat-sucrose (HFS) diet- induced nutritional obesity in rats.METHODS: A rat model of nutritional obesity was established by feeding rats a high-sugar and HFS diet. Obese rats were treated with TFGs for 8 weeks. During treatment, the weight of rats was recorded. At the end of treatment, the weight of epididymal fat pads was also mea- sured and fat tissue was taken for HE staining. Meanwhile, the levels of blood lipids, including serum triglyceride (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), and low density lipoprotein cholesterol (LDL-C), as well as fasting blood glucose (FBG) and fast- ing insulin (FINS), were determined. Homeosta- sis model assessment-insulin resistance (HOMA- IR) was calculated.RESULTS: Compared to rats fed a normal diet, rats fed a HFS diet had heavier body weight (495.5 g ± 69.2 g vs 388.7 g ± 60.2 g, P 〈 0.01) and epididymal fat pads (8.6 g ± 0.6 g vs 5.3 g ± 0.5 g, P 〈 0.01), larger adipocytes, increased levels of TG (0.9 mmol/L ± 0.3 mmol/L vs 0.3 mmol/L ± 0.2 mmol/L, P 〈 0.01), TC (2.7 mmol/L ± 0.4 mmol/L vs 1.5 mmol/L± 0.1 mmo1/L, P 〈 0.01), LDL-C (1.2 mmol/L ±0.2 mmol/L vs 0.5± 0.2 mmol/L, P 〈 0.01), FINS (27.8 ± 4.8 vs 13.6 ±3.6 mIU/L, P 〈 0.01), and HOMA-IR (8.0± 1.1 vs 3.5± 1.1, P 〈 0.01), and decreased HDL-C level (0.5 mmol/L±0.1 mmol/L vs 0.7 mmol/L± 0.2 mmol/L, P 〈 0.01). Treatment with TFGs caused a significant reduction in weight gain (462.5 g ± 55.2 g vs 495.5 g ± 69.2 g, P 〈 0.01), fat tissue accumulation (6.9 g ± 0.3 g vs 8.6 g ± 0.6 g, P 〈 0.01), TG (0.7 mmol/L ± 0.2 mmol/L vs 0.9 mmol/L ± 0.3 mmol/L, P 〈 0.01), TC (2.1 mmol/L ±0.3 mmol/L vs 2.7 mmol/L ± 0.4 mmol/L, P 〈 0.05), LDL-C (0.9 mrnol/L ±0.1 mmol/L vs 1.2 mmol/ L ± 0.2 mmol/L, P 〈 0.01), and HOMA-IR (5.1± 1.1 vs 9.2 ± 2.5,
